Branched-chain amino acids from plants and the metabolic syndrome: pathways and pharmacological applications

open access: yesFrontiers in Nutrition
BackgroundMetabolic syndrome (MetS) affects approximately 1.54 billion adults worldwide and is characterized by central obesity, insulin resistance, hypertension, and dyslipidemia. Chronic low-grade inflammation is central to the development of MetS.

Random generation of optimal saturated designs

open access: yes, 2013
Efficient algorithms for searching for optimal saturated designs are widely available. They maximize a given efficiency measure (such as D-optimality) and provide an optimum design. Nevertheless, they do not guarantee a \emph{global} optimal design. Indeed, they start from an initial random design and find a local optimal design.

Directed evolution of enzymes at the crossroads of tradition and innovation

open access: yesFEBS Open Bio, EarlyView.
An iterative cycle of data‐driven enzyme optimization comprising four stages: genetic diversification of a template enzyme, expression of protein variants, high‐throughput evaluation, and machine‐learning‐guided redesign of the next variant library.

Exploring the influence of lactoferrin on wound healing and related signaling pathways: a systematic review

open access: yesFEBS Open Bio, EarlyView.
Lactoferrin promotes wound healing by activating MAPK, PI3K/Akt, and extracellular matrix pathways, enhancing fibroblast remodeling, proliferation, and re‐epithelialization. These mechanisms highlight its therapeutic potential in inflammation control, tissue repair, and remodeling.

Optimizing photoexcitation conditions for time‐resolved X‐ray solution scattering experiments

open access: yesFEBS Open Bio, EarlyView.
Time‐resolved X‐ray solution scattering (TR‐XSS) is a powerful technique to visualize how proteins change their structure in real time after light activation. Selecting the right laser photoexcitation conditions—fluence, excitation geometry, and sample refresh rate—is critical to maximize the experimental signal while avoiding unwanted side effects ...

Saturated Fractional Design of Experiments: Toughness and Graphite Phase Optimizing in Nihard Cast Irons [PDF]

open access: yesJournal of Materials Engineering and Performance, 2007
The aim of the present research is to identify the manufacturing factors that exert an active influence on the graphite phase formation in mottled Nihard cast irons constituting the roll shells of duplex work rolls processed by the double pour method during centrifugal casting. The studied rolls, referred to as alloy indefinite chill, were processed at

Structural and biochemical insights into the thermostable esterase Ta0887 from Thermoplasma acidophilum

open access: yesFEBS Open Bio, EarlyView.
In this study, a novel esterase from the thermoacidophilic archaeon Thermoplasma acidophilum was biochemically and structurally characterized. Our results demonstrate that Ta0887 is a highly thermostable esterase that preferentially hydrolyzes p‐nitrophenyl hexanoate and possesses an α‐helical cap domain that likely contributes to its substrate ...

Unraveling 4‐Phenylbutyrate's Therapeutic Role in SLC6A1 Disorders: Pharmacochaperoning Over HDAC Inhibition

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objective Variants in SLC6A1, encoding the GABA transporter 1 (GAT‐1), cause epilepsy, autism spectrum disorder, and developmental delay via loss of GABA uptake, impaired trafficking, and ER retention. We previously found that 4‐Phenylbutyrate (PBA), an FDA‐approved drug, restores GABA uptake and reduces seizures in SLC6A1‐related disorders ...

Plasma EV Proteomics Identifies ECM Remodeling and Inflammatory Proteins LUM and C7 as Candidate Biomarkers in FSHD

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objective Facioscapulohumeral muscular dystrophy (FSHD) is one of the most debilitating and common muscular dystrophies. Despite its severity, no approved therapy exists for FSHD patients. However, several therapeutic candidates are currently under development, and some have recently entered clinical trials, marking the need for reliable ...

Optimal discrimination designs [PDF]

open access: yes
We consider the problem of constructing optimal designs for model discrimination between competing regression models. Various new properties of optimal designs with respect to the popular T-optimality criterion are derived, which in many circumstances ...
