Molecular Editing Reveals a Bilobalide Chemotype that Attenuates Prolyl Endopeptidase (PREP)‐Linked Neuroinflammation

open access: yesAdvanced Science, EarlyView.
Molecular editing of bilobalide uncovers BB56, a PREP‐engaging chemotype with enhanced anti‐inflammatory activity in microglia. Integrated CETSA‐MS, SAR, and pathway analyses link BB56 to attenuation of p38 MAPK/NF‐κB and NLRP3‐associated inflammatory signaling, highlighting bilobalide editing as a strategy for developing neuroinflammation‐modulating ...

A Generative Neuro‐Symbolic AI for Protein Sequence Design

open access: yesAdvanced Science, EarlyView.
We introduce EffieDes, a neuro‐symbolic framework coupling deep learning‐based fitness landscape parameterization with exact automated reasoning. Unlike greedy sampling, EffieDes identifies sequences that globally optimize fitness while satisfying intricate design constraints.

Artificial Host‐Guest Recognition Directs Glycometabolically Engineered Macrophages to Tumors

open access: yesAdvanced Science, EarlyView.
Glycoengineered supramolecular macrophages (GSAR‐M) are developed using glycometabolic labeling for bioorthogonal host‐guest tumor targeting. This engineering approach unexpectedly enhances macrophage migration, phagocytosis, and pseudopodia formation. Consequently, GSAR‐M demonstrate robust tumor‐targeting specificity, effectively arrest tumor growth,

Sustained Delivery of a Shingles Subunit Vaccine Overcomes Age‐Related Declines in Humoral and Cellular Immunity Relative to Shingrix

open access: yesAdvanced Science, EarlyView.
Sustained delivery of a shingles subunit vaccine from an injectable polymer–nanoparticle hydrogel enhanced the potency, durability, and breadth of immune responses in aged mice while minimizing inflammatory cytokine production and reactogenicity. This strategy decouples immunogenicity from reactogenicity and offers a promising approach to improve ...

A maritime search and rescue path planning method based on improved DQN

open access: yesInternational Journal of Digital Earth
In maritime search and rescue (SAR) missions, search path planning is a key factor in determining rescue efficiency and success rate. Due to the limitations of traditional algorithms – which rely heavily on manual decision-making, are strongly subjective,

An Improved Search and Rescue Algorithm for Global Optimization and Blood Cell Image Segmentation

open access: yesDiagnostics, 2023
Image segmentation has been one of the most active research areas in the last decade. The traditional multi-level thresholding techniques are effective for bi-level thresholding because of their resilience, simplicity, accuracy, and low convergence time,

Considerations for flight testing of search and rescue (SAR) innovations

open access: yesThe Journal of Search and Rescue
Flight testing of new aircraft, equipment and systems is mandatory in the aviation industry to establish and verify the airworthiness of any new airborne product as determined by the appropriate authority, for example the Federal Aviation Administration (FAA) in the United States or the European Union Aviation Safety Agency (EASA) in Europe.

From Displacement to Angle: Diamond‐Based 3D Rotation Sensing for High‐Precision Cellular Force Measurement

open access: yesAdvanced Science, EarlyView.
Cellular forces are traditionally inferred from displacement measurements, which lack rotational information and are constrained by linear model assumptions. This work replaces displacement with rotational angle for the fundamental mechanical readout.
