Human Engineered Heart Tissues Reproduce Trigger‐ and Substrate‐Driven Mechanisms of Ventricular Tachycardia

open access: yesAdvanced Science, EarlyView.
Engineered heart tissues (EHTs) derived from human iPSCs replicate mechanisms of ventricular tachycardia at the tissue level. Acquired Long‐QT conditions cause early afterdepolarizations, wavebreak, and transient rotors; in contrast, focal cryoinjury forms a stable substrate that supports ongoing reentry.

An Infusible Decellularized Extracellular Matrix Material Binds to Vasculature in Infarcted Myocardium and Induces Pro‐Reparative Gene Expression Following Acute Myocardial Infarction Through Inherent Avidity and Bioactive Signaling

open access: yesAdvanced Science, EarlyView.
To better characterize an infusible extracellular matrix (iECM) material, we applied size‐based fractionation to separate out components into two major fractions. We characterized these fractions to understand how material properties such as composition, morphology, and biochemical properties affect bioactive effects in the context of acute myocardial ...

NUB1 Regulates Muscle Atrophy in Lung Cancer Cachexia via NF‐κB Signaling Pathway

open access: yesAdvanced Science, EarlyView.
ABSTRACT Cancer‐associated cachexia (CAC) is a devastating multifactorial syndrome characterized by progressive skeletal muscle atrophy. Its mechanisms remain incompletely clarified, with limited effective therapeutic targets. NUB1, a negative regulator of ubiquitin‐like proteins mediating protein degradation, has not been fully elucidated in CAC ...

DUSP26 Regulated by YY1 Lactylation Couples Osteogenesis and Angiogenesis by Dephosphorylating PMPCB at Y141 and Activating Mitophagy in BMSCs

open access: yesAdvanced Science, EarlyView.
p300‐associated YY1 lactylation promotes DUSP26 expression in BMSCs. By regulating PMPCB phosphorylation and PINK1 processing, DUSP26 supports mitophagy, osteogenic differentiation, and pro‐angiogenic factor secretion. BMSC‐derived exosomes carrying a RUNX2 promoter‐driven DUSP26 plasmid favor osteogenic‐lineage transgene expression, partially ...

Loss of Lepr in Fibroblasts Drives Tumor Development by Promoting Macrophage Infiltration

open access: yesAdvanced Science, EarlyView.
TNFα and TGFβ signaling reprograms Lepr+ fibroblasts toward a LeprnegMmphigh inflammatory CAF (iCAF) state, with downregulation of the Tcf21‐Lepr axis contributing to this transition. Lepr loss induces Csf3, which contributes to the enhanced macrophage‐recruiting capacity of these iCAFs, thereby promoting tumor development. Thus, inflammatory signaling
