Bioadhesive Hydrogel With Polyphenol‐Armored Nanogene Rejuvenates Chondrocyte Senescence for Aged Osteoarthritis Therapy

An adhesive hydrogel depot with polyphenol‐armored nanogenes is developed for treating aged osteoarthritis. This system maintains miR‐140 bioactivity, prolongs its intra‐articular retention, and enhances transfection into chondrocytes, leading to the downregulation of senescence‐associated pathways.

Targeting a Myeloid–Regulatory B Cell Network Reverses Immune Paralysis in Periprosthetic Joint Infections

Periprosthetic joint infection establishes a sophisticated immunosuppressive network between CXCR4+ PMN‐MDSCs and Bregs, inducing profound CD8+ T cell paralysis. Alendronate effectively disrupts this CXCR4+ PMN‐MDSC–Breg axis by targeting STAT3, thereby restoring local immune surveillance.

THBS1+ Macrophages Exacerbate Modic Changes via SDC4‐Dependent Activation of NLRP3 Inflammasome

The illustration of THBS1+ macrophages exacerbate MCs via SDC4‐dependent activation of the NLRP3 inflammasome. THBS1+ macrophage subpopulation was identified in MCs through single‐cell RNA sequencing. C. acnes and its metabolites activate THBS1 expression in macrophages via the TLR signaling pathway.
