Eukaryotic serine racemase (SR) is a pyridoxal 5'-phosphate enzyme belonging to the Fold-type II group, which catalyzes serine racemization and is responsible for the synthesis of D-Ser, a co-agonist of the N-methyl-d-aspartate receptor. In addition to racemization, SR catalyzes the dehydration of D- and L-Ser to pyruvate and ammonia.

Serine palmitoyltransferase (SPT) is a key enzyme in sphingolipid biosynthesis and catalyzes the decarboxylative condensation of L-serine and palmitoyl coenzyme A to 3-ketodihydrosphingosine. We have succeeded in the overproduction of a water-soluble homodimeric SPT from Sphingomonas paucimobilis EY2395(T) in Escherichia coli.

Abstract The synthesis of O-(4,5-dimethoxy-2-nitrobenzyl)serine has been accomplished by treatment of an FMOC-aziridinecarboxylate with nitroveratryl alcohol followed by hydrolysis. Irradiation with a Pyrex-filtered 450W Hanovia lamp releases serine with a half-life of 4.4 min. o-Nitrobenzyl serinate is prepared by direct esterification.

KAHA Ligation at Serine

Oxazetidine assisted KAHA ligation was developed by the Bode group as an efficient peptide segment ligation strategy at native serine residues. This milestone achievement should enable the chemical synthesis of difficult-to-prepare proteins for biological studies.
