Disturbing Cholesterol/Sphingolipid Metabolism by Squalene Epoxidase Arises Crizotinib Hepatotoxicity

open access: yesAdvanced Science, Volume 12, Issue 14, April 10, 2025.
Crizotinib induces excessive accumulation of squalene epoxidase (SQLE) by blocking its autophagic degradation. This accumulation disrupts cholesterol and sphingolipid metabolism, leading to hepatic steatosis or hepatocyte apoptosis, respectively. Autophagy activators, such as metformin, or SQLE enzymatic inhibitors, like terbinafine, possesses the ...

Behavioral Abnormalities, Cognitive Impairments, Synaptic Deficits, and Gene Replacement Therapy in a CRISPR Engineered Rat Model of 5p15.2 Deletion Associated With Cri du Chat Syndrome

open access: yesAdvanced Science, Volume 12, Issue 14, April 10, 2025.
A new CdCS rat model is developed using CRISPR‐Cas9 technology. It shows impairments in cognitive and social behaviors, neuronal dendrite abnormalities in the hippocampus and mPFC, and inflammatory and immune processes in mPFC brain cells. Early AAV‐Ctnnd2 gene therapy enhances cognitive function in CdCS rats.

Escherichia coli in the production of biopharmaceuticals

open access: yesBiotechnology and Applied Biochemistry, Volume 72, Issue 2, Page 528-541, April 2025.
Abstract Escherichia coli has shouldered a massive workload with the discovery of recombinant DNA technology. A new era began in the biopharmaceutical industry with the production of insulin, the first recombinant protein, in E. coli and its use in treating diabetes. After insulin, many biopharmaceuticals produced from E. coli have been approved by the

Magnetic Resonance Imaging as a Readout of CLN5 Gene Therapy Efficacy in Sheep

open access: yesBrain and Behavior, Volume 15, Issue 4, April 2025.
Sheep with CLN5 Batten disease that received combination intracerebroventricular and intravitreal gene therapy underwent MRI scanning to assess global and regional brain volumes and determine the value of MRI to detect therapeutic efficacy. Sheep that were treated pre‐symptomatically underwent longitudinal scanning between 5 and 18 months of age, while

Lichen and Its Microbiome as an Untapped Source of Anti‐Biofilm Compounds

open access: yesChemistry &Biodiversity, Volume 22, Issue 4, April 17, 2025.
Lichens and their associated fungi give rise to the evaluation of several extracts and compounds against biofilms. ABSTRACT Lichen substances have been first described in the 1870s, and around 10 000 compounds have been isolated and characterized. Most of them have been evaluated for their activity on planktonic microorganisms (bacteria and fungi ...
