The Relationship between Rheumatoid “Factor” and Serum Desoxyribonucleic Acid

Summary A series of experiments undertaken to elucidate the identity of the rheumatoid “factor” present in the serum of rheumatoid arthritics have shown that this substance closely resembles desoxyribonucleic acid. In vitro DNA appears to complex with a Cohn IV-4 globulin and a Cohn Fraction III globulin.

Rheumatoid factors (RF) constitute the major autoantibodies in rheumatoid arthritis (RA). RF are directed against IgG Fc, are polyclonal, and are predominantly of the IgG and IgM classes. RF may participate in both synovial and extraarticular inflammation in RA, although the precise roles of serum IgG and IgM RF are unclear.

A simple radioactive binding assay for the detection of rheumatoid factor (RF-RBA) is described. Test sera are complement inactivated by incubation in 0.13 M ethylene diaminetetraacetic acid (EDTA) at 37 degrees C and then incubated with 125I-labelled heat-aggregated IgG.
