Definition and Discovery of Tandem SH3-Binding Motifs Interacting with Members of the p47phox-Related Protein Family

open access: yesBiomolecules
SH3 domains are widespread protein modules that mostly bind to proline-rich short linear motifs (SLiMs). Most known SH3 domain-motif interactions and canonical or non-canonical recognition specificities are described for individual SH3 domains.

Comparative assessment of crystallographic and cryo‐EM models in the Protein Data Bank

open access: yesFEBS Open Bio, EarlyView.
Raw data obtained by X‐ray crystallography or cryo‐EM result in experimental maps, ultimately fitted by atomic models. Although the physical principles are different, the final results can be viewed, compared, and evaluated in the same way. With cryogenic electron microscopy (cryo‐EM) on track to surpass X‐ray crystallography as the preferred method ...

Disordered but effective: short linear motifs as gene therapy targets for hyperexcitability disorders

open access: yesJournal of Clinical Investigation
Multiple approaches have targeted voltage-gated sodium (Nav) channels for analgesia. In this issue of the JCI , Shin et al. identified a peptide aptamer, Na V iPA1, carrying a short polybasic motif flanked by serine residues in a structurally disordered

Structural and biochemical insights into the thermostable esterase Ta0887 from Thermoplasma acidophilum

open access: yesFEBS Open Bio, EarlyView.
In this study, a novel esterase from the thermoacidophilic archaeon Thermoplasma acidophilum was biochemically and structurally characterized. Our results demonstrate that Ta0887 is a highly thermostable esterase that preferentially hydrolyzes p‐nitrophenyl hexanoate and possesses an α‐helical cap domain that likely contributes to its substrate ...

Optimising intrinsic disorder prediction for short linear motif discovery

open access: yes, 2019
Short linear motifs (SLiMs) are short protein regions, commonly only 3 - 10 amino acids in length, that are directly involved in protein-protein interactions. Identification of SLiMs is important for understanding fundamental processes involved in normal cellular function. SLiMs interact with their partnering proteins with low affinity. This makes them

The non-catalytic DNA polymerase ε subunit is an NPF motif recognition protein

open access: yesNature Communications
Short linear motifs (SLiMs) in disordered protein regions direct numerous protein–protein interactions, yet most remain uncharacterized. The Asn-Pro-Phe (NPF) motif is a well-known EH-domain ligand implicated in endocytosis, but here we reveal that the ...

Threonine 348 regulates the subcellular localization of PTEN

open access: yesFEBS Open Bio, EarlyView.
Thr348 in the C2 domain is a key contributor to PTEN subcellular localization. The PTEN350 fragment and PTENA4 accumulated in the nucleus, whereas PTENK13R,A4 predominantly localized to the plasma membrane. In contrast, substitution of Thr348 with Asp (T348D) disrupted these characteristic localization patterns, resulting in predominant cytoplasmic ...

A minimal cellulosome‐like system in Cellulosilyticum lentocellum

open access: yesFEBS Open Bio, EarlyView.
Cellulose‐degrading bacteria typically use cellulosomes, large multi‐enzyme complexes on a scaffold protein. In Cellulosilyticum lentocellum, we characterise a far smaller arrangement, a single scaffold bound to one cellulase through a single cohesin‐dockerin interaction.

BCG vaccination potentiates oxidative phosphorylation in neonatal myeloid‐derived suppressor cells

open access: yesFEBS Open Bio, EarlyView.
BCG vaccination enhances oxidative phosphorylation in neonatal MDSCs, impairing their immunosuppressive function. It upregulates electron transport chain genes and mitochondrial activity, increasing ATP and oxygen consumption. Pharmacological OXPHOS inhibition partially restores suppressive capacity, confirming causality.

The C‐terminal truncated splicing variant of NK1R negatively modulates substance P‐stimulated NK1R signaling

open access: yesFEBS Open Bio, EarlyView.
The neurokinin 1 receptor exists as full‐length (NK1L) and C‐terminally truncated (NK1S) splice variants. We show that NK1S heterodimerizes with NK1L, impairing Gαq coupling and Ca2+ mobilization while enhancing β‐arrestin1 recruitment. NK1S suppresses substance P‐driven gene expression and cell migration, revealing NK1S as an endogenous biased ...
