Multiplexed single molecule immunoassays

We have developed a method that enables the multiplexed detection of proteins based on counting single molecules. Paramagnetic beads were labeled with fluorescent dyes to create optically distinct subpopulations of beads, and antibodies to specific proteins were then immobilized to individual subpopulations.

We rarely get the opportunity to see how individual molecules behave. Mostly what we see are the average effects of large numbers of molecules. Molecular structures determined by crystallographic or NMR methods are time- and number-averages of a large population of molecules.
