目的 建立血清中佐匹克隆定性、定量测定的紫外分光光度法,为临床佐匹克隆中毒提供诊疗依据.方法 取0.5 ml血清加二氯甲烷2.0 ml充分震荡萃取,静置5 min,以4000 r/min离心,取二氯甲烷层于另一试管中氮气吹干,加0.1 mol/L盐酸溶液200μl混匀.取50μl于微量比色池中紫外扫描.结果 血清中佐匹克隆最大吸收峰(304±1) nm,浓度在1.0~30μg/ml范围内呈线性关系,回归方程y=0.0353x+0.0059,相关系数为0.9995,方法回收率为85.8%~102.5%;相对标准偏差(RSD)为:2.9%~4.5%.日内、日间RSD分别为2.73%~5.04%和2.97%~4.74%,最低检出浓度:0.5 μg/ml.结论 该方法操作简便、分析快速,结果准确 ...
