Engineering IL‐4 resistant proinflammatory human myeloid cells for cancer immunotherapy

open access: yesMolecular Oncology, EarlyView.
We developed a scalable workflow to generate proinflammatory human myeloid cells. CRISPR/Cas9‐edited CD34+ hematopoietic stem and progenitor cells were expanded and differentiated with M‐CSF. Deletion of STAT6 or STAT6/NFKB1 enhanced macrophage proinflammatory gene expression and cytokine secretion in the presence of IL‐4 while maintaining antibody ...

Stochastic Processes with Expected Stopping Time [PDF]

open access: yesLogical Methods in Computer Science
Markov chains are the de facto finite-state model for stochastic dynamical systems, and Markov decision processes (MDPs) extend Markov chains by incorporating non-deterministic behaviors.

Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

Stopping Criterion for Complexity Reduction of Sphere Decoding

open access: yesIEEE Communications Letters, 2009
Maximum-likelihood detection in MIMO communications amounts to solving a least-squares problem with a constellation (alphabet) constraint. One popular method that can be used to solve this problem is sphere decoding. We show in this letter that by employing a simple stopping criterion, it is possible to significantly reduce the complexity of sphere ...

Total Variation Wavelet-Based Medical Image Denoising

open access: yesInternational Journal of Biomedical Imaging, 2006
We propose a denoising algorithm for medical images based on a combination of the total variation minimization scheme and the wavelet scheme. We show that our scheme offers effective noise removal in real noisy medical images while maintaining sharpness ...

Versatile vector tools for efficient protein screening across multiple expression systems

open access: yesFEBS Open Bio, EarlyView.
A unified vector toolkit enables rapid protein expression screening across E. coli, insect, and mammalian cells. A single primer pair amplifies the target gene, which is inserted into any vector via a standardized interface. This streamlined workflow eliminates repeated cloning steps, accelerating the identification of optimal expression conditions for

Aquaporin‐3 and aquaporin‐5 impact the development of pancreatic ductal adenocarcinoma spheroids

open access: yesFEBS Open Bio, EarlyView.
Schematic representation of the role of aquaporin‐3 (AQP3) and aquaporin‐5 (AQP5) in pancreatic ductal adenocarcinoma (PDAC). Both proteins are upregulated in PDAC and are associated with tumor progression and metastatic potential. Silencing AQP3 or AQP5 in PDAC spheroids results in decreased diameter, area, and overall growth, underscoring their key ...

Electricity price prediction with univariate Long Short-Term Memory and zero-shot foundation transformer models

open access: yesEnergy and AI
Day-ahead electricity price forecasting remains challenging due to non-stationarity, extreme price spikes, and market-specific dynamics, while the potential of zero-shot foundation models relative to optimised supervised approaches remains underexplored.

Molecular characterization of covRS mutations in M1UK Streptococcus pyogenes

open access: yesFEBS Open Bio, EarlyView.
Group A Streptococcus (GAS) acquires covRS mutations driving a hypervirulent bacterial state, frequently associated with invasive disease‐like necrotizing fasciitis. We demonstrate that the newly emerged M1UK GAS lineage can also acquire these mutations.
