Engineering and Structural Elucidation of a Sac7d‐Derived IgG Fc‐Specific Affitin and Its Application for the Light‐Controlled Affinity Purification of Antibodies

open access: yesChemBioChem, EarlyView.
An affitin derived from the Sac7d scaffold enables the light‐controlled affinity purification of antibodies under physiological conditions, thus overcoming limitations of protein A chromatography. X‐ray crystallographic analysis and affinity measurements reveal complex formation with Fc at the CH2/CH3 junction, similar to protein A/G and FnRn. Combined

The Molecular Toolbox for Linkage Type‐Specific Analysis of Ubiquitin Signaling

open access: yesChemBioChem, EarlyView.
Ubiquitin modifications regulate virtually all aspects of eukaryotic cell biology through an array of polyubiquitin chain signals that adopt distinct structures, enabling the individual polyubiquitin linkages to mediate specific functions in cells.

Degradation of Nonribosomal Peptide Synthetase Megasynthetases SrfAA and SrfAB by Acyldepsipeptide‐Activated ClpP in Bacillus Subtilis

open access: yesChemBioChem, EarlyView.
FtsZ is the only target protein that has been biochemically confirmed to be hydrolyzed both in vitro and in bacterial cells. Coupled with activity‐based protein profiling of nonribosomal peptide synthetases, the ClpP‐ADEP1 proteolytic complex that degrades the surfactin synthetases SrfAA and SrfAB in cell lysates and bacterial cells is demonstrated ...

A Recognition Tag of Human Origin for Bioorthogonal Generation of Antibody‐Drug Conjugates using Microbial Biotin Ligase

open access: yesChemBioChem, EarlyView.
A human‐derived recognition tag of Pyrococcus horikoshii biotin ligase is fused to the monoclonal antibody trastuzumab at the C‐termini of either light or heavy chain. These fusion antibodies allow for the PhBL‐mediated introduction of desthiobiotin azide and the subsequent attachment of DBCO‐AF488 and DBCO‐Val‐Cit‐PAB‐MMAE via strain‐promoted azide ...
