This mini‐review describes the pathophysiology of the disrupted hypothalamic–pituitary–adrenal axis in dogs, and probably cats, with the MDR1 (ABCB1) mutation, ABCB1‐1Δ in dogs and ABCB11930‐1931del TC in cats.
