Substrate Gas Utilization and C3/C4 Metabolic Analysis of Actinobacillus succinogenes: Integration into a Model for Fermentation Prediction in BES

open access: yesFermentation
The aim of this work is to study the metabolism of Actinobacillus succinogenes in greater detail with the aim of optimizing succinate production and creating a metabolic model. The inhibitory properties of various substances were first investigated.

Effects of Molecular Designs and Double‐Network Morphologies for Bioadhesive Semiconductors

open access: yesAdvanced Functional Materials, EarlyView.
This study establishes molecular‐to‐mesoscale design rules for bioadhesive semiconducting polymers (BASCs). It identifies how side‐chain length, double‐network formation, and film thickness modulate adhesion strength and electronic performance, providing insight into the rational design of intrinsically adhesive semiconductors for stable and efficient ...

OXPHOS remodeling in high-grade prostate cancer involves mtDNA mutations and increased succinate oxidation

open access: yesNature Communications, 2020
Rewiring of energy metabolism and adaptation of mitochondria are considered to impact on prostate cancer development and progression. Here, we report on mitochondrial respiration, DNA mutations and gene expression in paired benign/malignant human ...

Microglial Fkbp5 Impairs Post‐Stroke Vascular Integrity and Regeneration by Promoting Yap1‐Mediated Glycolysis and Oxidative Phosphorylation

open access: yesAdvanced Science, EarlyView.
A post‐stroke perivascular niche of microglia characterized by low expression of M2 markers and elevated glycolysis, oxidative phosphorylation (OXPHOS), and phagocytic activity is identified, which is termed stroke‐activated vascular‐associated microglia (stroke‐VAM).

Succinate aggravates pulmonary fibrosis through the succinate/SUCNR1 axis

open access: yesAmerican Journal of Physiology-Lung Cellular and Molecular Physiology
This paper highlights the role of the succinate/SUCNR1 axis in pulmonary fibrosis. Receptor activation leads to profibrotic changes in IPF patient-derived fibroblasts. This finding could also be replicated in a mouse model of pulmonary fibrosis.

PTG‐Dependent Glycogen Metabolic Dysfunction Drives Impaired Adipose Browning: A Novel Mechanism Linking PM2.5 to Metabolic Disorders

open access: yesAdvanced Science, EarlyView.
This study provides the first evidence that PM2.5 impairs iWAT browning via PTG‐mediated glycogen metabolism disruption, which is initiated by ADRB3 inhibition and subsequently triggers VEGFB upregulation. It thereby delineates the ADRB3‐PTG‐VEGFB axis as central to PM2.5‐induced metabolic dysfunction and identifies adipose glycogen metabolism as a ...

Gut microbiota-derived succinate: Friend or foe in human metabolic diseases?

open access: yesReviews in Endocrine & Metabolic Disorders, 2019
There is now a wealth of evidence showing that communication between microbiota and the host is critical to sustain the vital functions of the healthy host, and disruptions of this homeostatic coexistence are known to be associated with a range of ...

Single‐Mitochondrion ATP Profiling Directs Discovery of Targetable OXPHOS Dependency in Cancers

open access: yesAdvanced Science, EarlyView.
MitoATP‐nFCM integrates nano‐flow cytometry with fluorogenic probes (ATP/membrane potential) and antibodies to quantify mitochondrial metabolites and protein expression at single‐organelle resolution, exposing oxidative phosphorylation (OXPHOS)‐driven metabolic rewiring in cancers.

Effects of Succinate on Growth Performance, Meat Quality and Lipid Synthesis in Bama Miniature Pigs

open access: yesAnimals
Succinate, one of the intermediates of the tricarboxylic acid cycle, is now recognized to play a role in a broad range of physiological and pathophysiological settings, but its role in adipogenesis is unclear.

Caspase‐3/GSDME‐Mediated Trophoblast Pyroptosis and Reciprocal Macrophage Polarization Contribute to Inflammation in Early‐Onset Preeclampsia

open access: yesAdvanced Science, EarlyView.
In trophoblast cells, T/S (TNFα + SM164) activated caspase‐3, which cleaved GSDME to switch apoptosis to pyroptosis, causing cell swelling, membrane rupture, and release of LDH, HMGB1, IL‐1β and IL‐18. These cytokines induced pro‐inflammatory macrophage polarization, which in turn reinforced pyroptotic signaling in trophoblasts, amplifying systemic ...
