1932 entdeckte Domagk die antibakterielle Wirkung des ersten systemisch anwendbaren Chemotherapeuticums, des von Mietzsch und Klarer hergestellten Azofarbstoffes Prontosil, der eine Sulfon-amidgruppe besitzt. 1935 zeigen Trefouel u. Mitarb., das das in vivo aus Prontosil entstehende Sulfanilamid der eigentlich wirksame Molekulanteil ist.
