A 3D Human Bone and Bone Marrow‐on‐a‐Chip Model for In Vitro Bone Remodeling and Immune Cell Maintenance
Advanced Science, EarlyView. This study presents a human Bone (and Bone Marrow)‐on‐a‐Chip model based on native human bone scaffolds and autologous cells. Dynamic perfusion and sequential cell seeding replicate the physiological bone remodeling process in vitro, enabling the long‐term culture of functional, mature bone marrow immune subpopulations.Nina Stelzer, Melanie‐Jasmin Ort, Kristian Händler, Emely Bortel, Ioanna Maria Dimitriou, Martin Textor, Georg N. Advanced Science, EarlyView.Bone cancer pain and depression share a common origin: astrocytic A2‐to‐A1 transition in the posterior piriform cortex. This phenotypic shift disrupts the ATP–adenosine–A2AR–norepinephrine axis, simultaneously driving nociceptive and affective dysfunction.Jiang‐Ping Liu, Jun‐Han Zhang, Zhi‐Xuan Tan, Wei‐Bing Yan, Ya‐Ru Yuan, Yao Liu, Bing‐Di Wei, Meng‐Qi Ding, Zhuo‐Min Fu, Ying Liu, An‐Qi Wang, Lin Ma, Yong‐Hong Li, Er‐Qing Chai, Chao‐Jun Wei +14 morewiley +1 more sourceCysteine Redox State Governs the Condensation Pathway of Hendra Virus W Protein and Differentially Impacts Type I IFN and NF‐κB Signaling
Advanced Science, EarlyView. The cysteine redox state of the Hendra virus W (WHeV) protein controls assembly into fibrils or amorphous aggregates, with residues 2–29 required for fibrillation. In cells, WHeV forms different types of nuclear condensates. Impaired ability to form redox‐sensitive, non‐filamentous condensates reduces WHeV inhibition of the NF‐κB pathway, while it ...Frank Gondelaud, Alexandre Lalande, Giulia Pesce, Auriane Bouchet, Carole Meunier, Christophe Bignon, Denis Ptchelkine, Yu Gu, Eva Ogire, Pierre‐Yves Lozach, Denis Gerlier, Cyrille Mathieu, Sonia Longhi +12 morewiley +1 more sourceMultimodal Imaging Reveals Rapid Catecholamine Uptake and Release by Neutrophils
Advanced Science, EarlyView. We show that immune cells (neutrophils) synthesize, uptake, and store catecholamine neurotransmitters such as dopamine or adrenaline. They also release them in response to specific stimuli (serotonin), which we directly visualize using fluorescent nanosensors. We further demonstrate that catecholamines affect neutrophil functions (NETosis) and platelet Jennifer Mohr, Anne Schmitz, Meshkat Dinarvand, Franziska Wulfert, Sangeetha Shankar, Bjoern F.
