Purification and characterization of a liposomal-forming tetraether lipid fraction

Biochemical and Biophysical Research Communications, 1990
Polar lipid E, PLE, a native tetraether lipid mixture from Sulfolobus acidocaldarius is shown to spontaneously form multilamellar liposomes in aqueous media. PLE lipids were isolated as a single fraction from the crude lipid extract of S. acidocaldarius on a reverse-phase column followed by TLC and methanol precipitation.

Archaebacterial bipolar tetraether lipids: Physico-chemical and membrane properties

Chemistry and Physics of Lipids, 2010
Bipolar tetraether lipids (BTL) are abundant in archaea and can be chemically synthesized. The structures of BTL are distinctly different from the lipids found in bacteria and eukaryotes. In aqueous solution, BTL can form extraordinarily stable liposomes with different sizes, lamellarities and membrane packing densities.

Calditol tetraether lipids of the archaebacterium Sulfolobus solfataricus. Biosynthetic studies.

The Biochemical journal, 1990
Lipids from the archaebacterium Sulfolobus solfataricus are based on 72-membered macrocyclic tetraethers made up from two C40 diol units differently cyclized and either two glycerol moieties or one glycerol moiety and a unique branched-chain nonitol named calditol (glycerodialkylnonitol tetraethers, GDNTs). To elucidate the biosynthesis of calditol and

Synthetic Studies of Archaeal Macrocyclic Tetraether Lipids: Practical Synthesis of 72-Membered Tetraether Model Compounds

Bulletin of the Chemical Society of Japan, 1997
Abstract Archaeal 72-membered macrocyclic tetraether model compounds (5 and 6) were synthesized by intramolecular dicarbonyl coupling with the aid of low-valent titanium, which is known as McMurry coupling. This synthetic methodology is the first practical way to obtain the two regioisomeric structures in quantity and appears to be ...

Stability and proton-permeability of liposomes composed of archaeal tetraether lipids

Biochimica et Biophysica Acta (BBA) - Biomembranes, 1994
Liposomes composed of tetraether lipids originating from the thermoacidophilic archaeon Sulfolobus acidocaldarius were analyzed for their stability and proton permeability from 20 degrees C up to 80 degrees C. At room temperature, these liposomes are considerably more stable and have a much lower proton permeability than liposomes composed of diester ...

Dynamics and Organization of Archaeal Tetraether Lipid Membranes

2017
Archaeal bipolar tetraether lipids (BTLs) have distinct structural differences from the lipids isolated from bacteria and eukaryotes. Because of the presence of the unusual structural features, such as macrocyclic structures, cyclopentane rings, isoprenoid units, tetraether linkages, and a variety of polar head groups, archaeal BTL membranes possess ...

Unusual Thermal Stability of Liposomes Made from Bipolar Tetraether Lipids

Biochemical and Biophysical Research Communications, 1994
The thermal stability of liposomes made from polar lipids of Sulfolobus acidocaldarius, a thermoacidophilic archaebacterium, was studied. While two analogous nonarchaebacterial liposomes examined were unstable above 50 degrees C, liposomes made of the native tetraether lipids showed stability up to at least 80 degrees C.

Selective interactions of concanavalin A‐modified tetraether lipid liposomes

physica status solidi c, 2011
AbstractFor the development of site specific liposomes, high stability and long circulation properties are very promising for the improvement of drug targeting and drug delivery system as well as improvement of bioavailability of efficient bioactive drugs.

Capillary supercritical fluid chromatography of archaebacterial glycerol tetraether lipids

Journal of High Resolution Chromatography, 1986
AbstractThe analysis of bacterial glycerol tetraethers by capillary supercritical fluid chromatography is described. Complete separation of ethers, differing only by the number of cyclopentane rings present in the isopranyl side chains could be achieved.
