5,10-Methylene-tetrahydrofolate reductase (MTHFR) deficiency is a rare, autosomal recessive, metabolic disorder of folate metabolism, which affects homocysteine remethylation. Elevated homocysteine with normal or low methionine level is the key to diagnosis.

Oxidative degradation of dihydrofolate and tetrahydrofolate

7,8-Dihydrofolate has been degraded oxidatively using ferricyanide under anaerobic conditions. Three reaction pathways have been found for this oxidation, and these pathways can be controlled by variation of the pH and by the nature of the buffer anion.

5-Methyltetrahydrofolate and vitamin B12 appear to be required for the biosynthesis of tetrahydrobiopterin. A deficiency of either could be sufficient to bring about neurological change which can be corrected by reversing the deficiency. Patients with senile dementia could possibly be benefited by the administration of 5-methyltetrahydrofolate.

Abstract A survey of fluorescence characteristics of tetrahydrofolate and related compounds has been carried out with an Aminco-Bowman Spectrophotofluorometer. Dihydrofolate and all the tetrahydrofolate derivatives examined, except 5,10-methenyltetrahydrofolate, show maximum excitation between 300 to 320 mμ and maximum fluorescence between 360 to 425
