A General Three-Component Alkyl Petasis Boron-Mannich Reaction
Aryl amines are one of the most common moieties in biologically active molecules, and approximately 37% of drug candidates contain aromatic amines. Recent advancements in medicinal chemistry, coined as “escaping from flatland”, have led to a greater focus on accessing highly functionalized C(sp3)-rich amines to improve the physiochemical and ...

This study characterizes the responses of primary acute myeloid leukemia (AML) patient samples to the MCL‐1 inhibitor MIK665. The results revealed that monocytic differentiation is associated with MIK665 sensitivity. Conversely, elevated ABCB1 expression is a potential biomarker of resistance to the treatment, which can be overcome by the combination ...

Aggressive prostate cancer is associated with pericyte dysfunction
Tumor‐produced TGF‐β drives pericyte dysfunction in prostate cancer. This dysfunction is characterized by downregulation of some canonical pericyte markers (i.e., DES, CSPG4, and ACTA2) while maintaining the expression of others (i.e., PDGFRB, NOTCH3, and RGS5).

Survivin and Aurora Kinase A control cell fate decisions during mitosis
Aurora A interacts with survivin during mitosis and regulates its centromeric role. Loss of Aurora A activity mislocalises survivin, the CPC and BubR1, leading to disruption of the spindle checkpoint and triggering premature mitotic exit, which we refer to as ‘mitotic slippage’.

Intein‐based modular chimeric antigen receptor platform for specific CD19/CD20 co‐targeting
CARtein is a modular CAR platform that uses split inteins to splice antigen‐recognition modules onto a universal signaling backbone, enabling precise, scarless assembly without re‐engineering signaling domains. Deployed here against CD19 and CD20 in B‐cell malignancies, the design supports flexible multi‐antigen targeting to boost T‐cell activation and

Puzzling three-component disorder from unprecedented reaction
The synthesis of an iron nitrilimido complex results in a mixture of products, which have similar size requirements. A co-crystal of two products, in addition to the residual starting material from the prior reaction, features a structure with a challenging disorder of different iron complexes occupying the same site.
