Analysis of Actin Assembly by In Vitro TIRF Microscopy

Methods in Molecular Biology, 2009
Since directed movement toward an extracellular chemoattractant requires rapid and continuous reorganization of the actin cytoskeleton to form complex structures such as a protruding lamellipodium, it is of great interest to analyze and understand the individual contribution of proteins specifically involved in this process.

Localization and classification of membrane dynamics in TIRF microscopy image sequences

open access: yes2014 IEEE 11th International Symposium on Biomedical Imaging (ISBI), 2014
The detection of proteins and the classification of their temporal behaviors in live cell fluorescence microscopy are of utmost importance to understand cell mechanisms. In this paper, we aim at locating and recognizing temporal events in TIRF microscopy image sequences related to membrane dynamics.

Single Molecule Genotyping by TIRF Microscopy

Journal of Fluorescence, 2008
As part of a programme to develop a metrological framework for single molecule measurements in biology, we have investigated the applications of single molecule imaging to genomics. Specifically, we have developed a technique for measuring the frequencies of single nucleotide polymorphisms (SNPs) in complex or pooled samples of DNA.

TIRF

Imaging & Microscopy, 2009
Changes in the intracellular Ca2+ concentration ([Ca2+]i) accompany many diverse biological phenomena from neuronal excitability and muscle contraction to activation of transcription and cell death. Thanks to the introduction of fluorescent intracellular Ca2+ dyes our understanding of Ca2+ signalling has greatly advanced; it is established that ...

Antibody immobilisation on fibre optic TIRF sensors

Biosensors and Bioelectronics, 2003
A study of antibody immobilisation techniques on quartz and fibre optic surfaces for immunosensors has been carried out. Methods of covalent antibody immobilisation which have not previously been applied to optical fibres were investigated, and compared with classical methods found in the literature.

Competitive Protein Adsorption Studied with TIRF and Ellipsometry

open access: yesJournal of Colloid and Interface Science, 1996
Total internal reflection fluorescence (TIRF) and ellipsometry have been used to study competitive protein adsorption to a hydrophobic model surface prepared by radio frequency plasma deposition of hexamethyl disiloxane on silicon.

Confocal TIRF microscopy of single molecules

SPIE Proceedings, 2003
Total internal reflection fluorescence (TIRF) microscopy is a powerful technique to investigate surface bound emitters exclusively, i.e. with low interference of the fluorescing bulk in the adjacent aqueous solution. Confocal microscopy is a powerful technique to detect low emission intensities, like the emission of a single molecule, with high ...

Surface Plasmon Enhanced TIRF Imaging

Imaging & Microscopy, 2009
Total Internal Reflection Fluorescence Microscopy (TIRFM) is a powerful technique for imaging dynamic membrane events in living cells. However the information gained from TIRFM may be limited by the intensity of the florescence signal and by background noise emanating from the inner part of the cell.

Monitoring fibrinogen adsorption kinetics by interfacial TIRF rheometry

Journal of Molecular Recognition, 1996
The adsorption kinetics of purified fibrinogen to unmodified and aminopropylsilane-modified quartz glass surfaces were studied under pseudo-first order (binding-unit excess) conditions by the total internal reflection fluorescence (TIRF) method. Fluorescence in the adsorbed protein layer (350 nm) was excited by the evanescent wave at 285-290 nm.

Microtubule Dynamics at the Cell Cortex Probed by TIRF Microscopy

2010
Total internal reflection fluorescence (TIRF) microscopy is a technique that allows selective excitation of fluorescence at a liquid/solid interface within a short distance from the boundary. The penetration depth of TIRF microscopy depends on the angle of illumination resulting in a range of depths, which typically vary from approximately similar 70 ...
