Molecular phylogeny of miR390-guided trans-acting siRNA genes (TAS3) in the grass family
Trans-acting siRNAs (tasiRNAs) are a plant-specific class of 21-nt endogenous siRNAs that function as miRNA-like posttranscriptional negative regulators. Several tasiRNA loci (known as TAS genes) have been characterized to date in rice and Arabidopsis. The TAS3 family is distinguished from other TAS loci by the dual miR390 complementary sites flanking ...

To endure considerable fluctuations in temperature, plants need precise regulation of temperature-controlled gene expression. In this study, the involvement of TAS1 trans-acting siRNA (tasiRNA) in temperature-controlled gene expression was examined in Arabidopsis. The accumulation of TAS1 tasiRNA was downregulated at 4 degrees C.

Arabidopsis thaliana encodes four Dicer-like (DCL) proteins and five dsRNA-binding (DRB) proteins. We have previously demonstrated that DCL4 specifically interacts with DRB4 in vitro. Here we describe the interaction between DCL4 and DRB4 in vivo. The phenotype of a mutant with a defect in DCL4 (dcl4-2) was similar to that of a mutant with a defect in ...

Roles of target site location and sequence complementarity in trans‐acting siRNA formation in Arabidopsis [PDF]
SummaryIn plants, many mRNAs and non‐coding RNAs are cleaved by RNA‐induced silencing complexes. After cleavage, only a limited number of RNAs are processed into trans‐acting siRNAs (tasiRNAs). One reason is that 22 nt small RNAs, but not the more common 21 nt small RNAs, can efficiently trigger tasiRNA formation.

MicroRNA-Dependent Trans-Acting siRNA Production
Science's STKE, 2005Less than a year has elapsed between the discovery of trans-acting small interfering RNAs (tasiRNAs) in plants and the elucidation of the major steps of the corresponding pathway. During tasiRNA biogenesis, polyadenylated RNAs transcribed from non–protein-coding TAS genes are cleaved by a microRNA (miRNA)–programmed
