Treating patients with low high-density lipoprotein cholesterol: choices, issues and opportunities

open access: yesCurrent Controlled Trials in Cardiovascular Medicine, 2001
Three clinical trials have recently focused on the benefits of lipid-regulating therapy in populations with normocholesterolaemia and low high-density lipoprotein (HDL)-cholesterol.

A Super‐Enhancer‐Driven Transcriptional Regulatory Circuit Underlying Abiraterone Resistance in Castration‐Resistant Prostate Cancer

open access: yesAdvanced Science, EarlyView.
This study identifies a super‐enhancer‐driven transcriptional regulatory circuit comprising BCL6, SMAD3, and NFIB that cooperate to drive cholesterol synthesis via SREBF2/HMGCR/FDFT1 activation and regulate CDK2/CCND3 for cell cycle control. Targeting this circuit with BI‐3802 or downstream inhibitors (Fatostatin/Lovastatin) overcomes abiraterone ...

Metformin Lowers Plasma Triglycerides by Promoting VLDL-Triglyceride Clearance by Brown Adipose Tissue in Mice

open access: yesDiabetes, 2014
Metformin is the first-line drug for the treatment of type 2 diabetes. Besides its well-characterized antihyperglycemic properties, metformin also lowers plasma VLDL triglyceride (TG).

RPL6 Interacts with HMGCS1 to Stabilize HIF‐1α by Promoting Cholesterol Production in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
The study found that RPL6 is frequently upregulated in HCC tissues with extrahepatic metastasis. Elevated RPL6 bound to and increased the stability of HMGCS1 mRNA, thereby enhancing intracellular cholesterol levels. Subsequently, increased cholesterol promotes cancer metastasis in HCC by stabilizing HIF‐1α protein.

The TET3/GATA6 Axis Drives Lipid Metabolism and Therapeutic Vulnerabilities in Pancreatic Ductal Adenocarcinoma

open access: yesAdvanced Science, EarlyView.
The DNA demethylase TET3 drives lipid metabolic reprogramming in pancreatic ductal adenocarcinoma via a non‐catalytic mechanism. TET3 recruits histone deacetylases to repress GATA6, sustaining lipogenic enzyme expression and ferroptosis resistance.

Central lipid detection and the regulation of feeding behavior

open access: yesOléagineux, Corps gras, Lipides, 2013
The modern abundance of energy-rich foods combined with a shift to more sedentary lifestyles has led to a thermodynamic imbalance in which excessive caloric intake and reduced energy expenditure account for the prevalence of obesity.
