Intracellular trafficking of tropoelastin

Matrix Biology, 1998
Elastin is secreted as soluble tropoelastin monomers which are then cross-linked in the presence of extracellular microfibrils to form insoluble elastic fibers. Although the secretion of tropoelastin is thought to be mediated and targeted by an intracellular chaperone complex, the intracellular route taken by this protein and the role of such a ...

Proteolysis of Tropoelastin

1977
The presence of an enzyme associated with tropoelastin is described. The enzyme has a pH optimum between 7 and 9 and trypsin-like specificity. Upon incubation, tropoelastin (72,000 molecular weight) is cleaved into relatively high molecular weight fragments. In addition to the parent molecule, five discrete polypeptide bands are usually observed on SDS

Polyclonal antibodies to tropoelastin and the specific detection and measurement of tropoelastinin vitro

Connective Tissue Research, 1991
Because tropoelastin is difficult to purify, most antibodies to elastin are raised against the insoluble form of the molecule. While these antibodies cross-react with tropoelastin, antigenic differences between insoluble and soluble elastin suggest that antibodies raised directly against tropoelastin might provide a more sensitive and specific reagent ...

Tropoelastin Biosynthesis by Corneal Cells

Ophthalmologica, 2003
The vertebrate cornea is an avascular tissue and does not contain elastic fibers. We tested the capacity of corneal epithelial cells and stromal keratocytes to synthesize tropoelastin. Explant cultures and cell cultures were obtained from these two cell types in standard culture conditions.

Biosynthesis of Tropoelastin by Elastic Cartilage

Connective Tissue Research, 1980
Elastic ear cartilage and aortae from nine-day-old hamsters were incubated in Krebs-Ringer medium containing ascorbate, beta-aminopropionitrile and either [14C]proline or [3H]valine. The [14C]proline-labeled proteins were separated by chromatography on Agarose A-5 m in sodium dodecyl sulfate and their [14C]-hydroxyproline content measured.

Tropoelastin Expression by Periodontal Fibroblasts

Journal of Dental Research, 2002
Elastic system fibers are load-bearing proteins found in periodontal tissue. There are three types—oxytalan, elaunin, and elastic fibers—which differ in their relative microfibril and elastin contents. Oxytalan fibers are known to be distributed in the periodontal ligaments and gingiva, whereas elaunin and elastic fibers are present only in the ...

Multiple chick tropoelastin mRNAs

Biochemical and Biophysical Research Communications, 1988
Several overlapping chick tropoelastin cDNAs were isolated from a lambda gt11 cDNA library constructed from whole 10 day chick embryo total RNA. Comparison of the nucleotide sequence of the 2.3 kb tropoelastin cDNA to the sequences published by Bressan et al. (1) and Tokimitsu et al.

Characterization of rat heart tropoelastin

Archives of Biochemistry and Biophysics, 1989
Several overlapping rat tropoelastin cDNA clones were isolated from a lambda gt11 rat heart cDNA library and their nucleotide sequence was determined. The corresponding deduced amino acid sequence of rat tropoelastin revealed strong homology to bovine and human tropoelastins although possessing some unique features including greater size (18%) and ...

[44] Preparation of antiserum to tropoelastin

1982
Publisher Summary In preparation of antiserum, normally three rabbits weighing approximately 3 kg are used to ensure adequate antiserum titers. Purified tropoelastin is dissolved in Na 2 HPO 4 , NaC1, pH 7.4 and emulsified in an equal volume of Freund's complete adjuvant.

Primary Structure of Porcine Tropoelastin

1977
Structural studies on porcine aortic tropoelastin as obtained from copper-deficient swine have been performed on the whole protein as well as fragments produced by trypsin, subtilisin and thrombin digestion. Tryptic fragments fall into two catagories, those large, over 20 residues, and those small, 4 residues and less.
