Primary Structure of Porcine Tropoelastin

Structural studies on porcine aortic tropoelastin as obtained from copper-deficient swine have been performed on the whole protein as well as fragments produced by trypsin, subtilisin and thrombin digestion. Tryptic fragments fall into two catagories, those large, over 20 residues, and those small, 4 residues and less.

Elastic ear cartilage and aortae from nine-day-old hamsters were incubated in Krebs-Ringer medium containing ascorbate, beta-aminopropionitrile and either [14C]proline or [3H]valine. The [14C]proline-labeled proteins were separated by chromatography on Agarose A-5 m in sodium dodecyl sulfate and their [14C]-hydroxyproline content measured.

Fibrillation of tropoelastin induced by proteoglycan

Abstract The true monomerix size of porcine aortic tropoelastin is 72,000–74,000 daltons. The newly modified isolation procedure we describe gives meterial predominantly of this molecular size with improved yields. The protein is extremely susceptible to proteolysis and steps must be taken during isolation to prevent this occurrence.
