The full-length TSH receptor is stabilized by TSH ligand
Journal of Molecular Graphics and Modelling, 2023The receptor for thyroid stimulating hormone (TSHR), a GPCR, is the primary antigen in autoimmune hyperthyroidism (Graves' disease) caused by stimulating TSHR antibodies. While we have previously published a full length model of the TSHR, including its leucine rich domain (LRD), linker region (LR) and transmembrane domain (TMD), to date, only a partial

Abstract. When solubilized, radiolabelled membrane preparations from FRTL-5 rat thyroid cells are applied to TSH affinity columns, two separate peaks of protein can be eluted by high salts/high pH and low pH buffers, respectively. Immunoprecipitation with monoclonal antibodies to the TSH receptor shows that both peaks contain proteins related to the ...

A radioligand receptor assay (RRA) for TSH and TSH-binding-inhibiting antibodies (TBIAb) using solubilized receptor is presented. Introducing charcoal for separation of bound and free hormone has made the assay easier to handle. The detection limit for TSH is 30 microU/ml as measured by RIA. TBIAb was found in the sera of 19 of 26 hyperthyroid patients.

Recombinant TSH-Receptor for Determination of TSH-Receptor-Antibodies
Experimental and Clinical Endocrinology & Diabetes, 2009We have expressed the complete coding sequence of the human TSH-R in eucaryotic cells. Limiting dilution enabled us to select two clones, JP09 and JP26, which have formed the basis of binding and bioassays respectively, for TSH-R antibodies. Results obtained in the binding assay correlated well with those obtained in the TRAK (Henning Berlin) assay ...
