Development of therapies targeting cancer‐associated fibroblasts (CAFs) necessitates preclinical model systems that faithfully represent CAF–tumor biology. We established an in vitro coculture system of patient‐derived pancreatic CAFs and tumor cell lines and demonstrated its recapitulation of primary CAF–tumor biology with single‐cell transcriptomics ...

Breast cancer or metastasis? An unusual case of metastatic malignant pleural mesothelioma to the breast [PDF]
Background: Metastases to the breast from extramammary malignancies are very rare, and ruling out the diagnosis of primary breast tumor is important in order to decide on clinical management and predict prognosis.

Dual targeting of RET and SRC synergizes in RET fusion‐positive cancer cells
Despite the strong activity of selective RET tyrosine kinase inhibitors (TKIs), resistance of RET fusion‐positive (RET+) lung cancer and thyroid cancer frequently occurs and is mainly driven by RET‐independent bypass mechanisms. Son et al. show that SRC TKIs significantly inhibit PAK and AKT survival signaling and enhance the efficacy of RET TKIs in ...

In vitro evidence for senescent multinucleated melanocytes as a source for tumor-initiating cells [PDF]
Oncogenic signaling in melanocytes results in oncogene-induced senescence (OIS), a stable cell-cycle arrest frequently characterized by a bi- or multinuclear phenotype that is considered as a barrier to cancer progression.

The noncoding region of the genome plays a key role in regulating gene expression, and mutations within these regions are capable of altering it. Researchers have identified multiple functional noncoding mutations associated with increased cancer risk in the genome of breast cancer patients.

We developed and validated a DNA methylation–based biomarker panel to distinguish pleural mesothelioma from other pleural conditions. Using the IMPRESS technology, we translated this panel into a clinically applicable assay. The resulting two classifier models demonstrated excellent performance, achieving high AUC values and strong diagnostic accuracy.

Chromosomal Gains and Losses in Uveal Melanomas Detected by Comparative Genomic Hybridization [PDF]
Eleven uveal melanomas were analyzed using comparative genomic hybridization (CGH). The most abundant genetic changes were loss of chromosome 3, overrepresentation of 6p, loss of 6q, and multiplication of 8q.

The DNA methylation inhibitor 5-aza-2’-deoxycytidine retards cell growth and alters gene expression in canine mammary gland tumor cells [PDF]
Disruption of gene expression by DNA methylation changes is widely involved in tumorigenesis. Here,to investigate DNA methylation changes in canine, we treated a canine mammary gland tumor cell line with a DNA methylation inhibitor, 5-aza-2 ...
