Von In‐Silico bis In‐Cerebro – Entwicklung des Orexinrezeptor Antagonisten „Photorexant“ zur Photomodulation In Vitro und im Gehirn der Maus

Durch einen strukturbasierten Designansatz wurden photoschaltbare Derivate des von der FDA zugelassenen Arzneistoffes Suvorexant entwickelt, gedockt, synthetisiert und pharmakologisch an den Orexin‐1‐ und 2‐Rezeptoren (OX1R und OX2R) charakterisiert.

Lichtaktivierbare Iridium(III)‐Prodrugs zeigen, wie die Koordination von Chalkogenen die photoinduzierte therapeutische Aktivität steuert. Bei Bestrahlung mit sichtbarem Licht laufen an den schwefel‐ bzw. selenhaltigen Liganden unterschiedliche, sauerstoffabhängige Photoreaktionen ab.

Aufgrund überlagerter oder konkurrierender Mechanismen sind relevante Wechselwirkungen bei der Staubung von Schüttgutmischungen komplex und nur unzureichend verstanden. Selbst für Mischungen aus umfassend charakterisierten Einzelkomponenten existieren keine belastbaren Modelle. In dieser Arbeit werden die Hürden bei der Prognose thematisiert.
