UCHL3 Regulates Subgenomic Flaviviral RNA Condensates to Promote Virus Propagation

open access: yesAdvanced Science, EarlyView.
ABSTRACT Flavivirus subgenomic RNAs (sfRNAs) antagonise antiviral defences, yet how sfRNAs are organized and maintained in cells remains poorly understood. Here we identify ubiquitin C‐terminal hydrolase L3 (UCHL3) as a post‐translational regulator of flavivirus sfRNA stability and function.

Decoys provide a scalable platform for the identification of plant E3 ubiquitin ligases that regulate circadian function

open access: yeseLife, 2019
The circadian clock relies on regulated degradation of clock proteins to maintain rhythmicity. Despite this, we know few components that mediate protein degradation.

Unveiling Lactylation: A Novel Frontier in Cancer Stemness and Therapy

open access: yesAdvanced Science, EarlyView.
Tumor metabolism and epigenetic regulation are intrinsically linked. This review highlights lactylation as a pivotal bridge connecting these processes, acting as an active epigenetic effector in cancer stemness. We delineate how lactylation orchestrates the cancer stem cell landscape and evaluate its regulatory networks.

Auto‐Downregulation of the Florigen FT Production Prevents Precocious Flowering in Plants

open access: yesAdvanced Science, EarlyView.
This study shows that the leaf vein‐expressed FD complexes with the florigen FT to bind DNA motifs in FT promoter, which antagonizes CONSTANS‐mediated FT activation to feedback down‐regulate FT expression. This prevents an excessive induction of FT expression by inductive photoperiods, thereby precluding precocious transition to flowering in ...

Structures of Two Melanoma-Associated Antigens Suggest Allosteric Regulation of Effector Binding [PDF]

open access: yes, 2016
The MAGE (melanoma associated antigen) protein family are tumour-associated proteins normally present only in reproductive tissues such as germ cells of the testis.

Structural ubiquitin contributes to K48 linkage specificity of the HECT ligase Tom1

open access: yesCell Reports
Summary: Homologous to E6AP C terminus (HECT) ubiquitin ligases play key roles in essential pathways such as DNA repair, cell cycle control, or protein quality control. Tom1 is one of five HECT ubiquitin E3 ligases in budding yeast S.

Targeting METTL3 Attenuates Thyroid Inflammatory Injury by Restoring Th17/Treg Balance through a YTHDC2‐m6A‐Dependent KDR/VEGFA Loop

open access: yesAdvanced Science, EarlyView.
METTL3‐mediated m6A modification stabilizes KDR/Kdr mRNA through YTHDC2, amplifying the VEGFA‐KDR feedback loop and disturbing Th17/Treg balance. This pathway promotes persistent inflammation and tissue damage in patients with AIT, and inhibiting the METTL3‐KDR axis effectively disrupts this circuit, alleviating thyroid tissue injury and disease ...

Role of HECT ubiquitin protein ligases in Arabidopsis thaliana [PDF]

open access: yes, 2018
Ubiquitination is a kind of posttranslational modification of proteins in eukaryotes, and it plays an important role in the growth and development of organisms. The ubiquitination of proteins is a cascade enzymatic reaction involving three enzymes.

Ubiquitin Ligases in Control: Regulating NLRP3 Inflammasome Activation

open access: yesFrontiers in Bioscience-Landmark
Ubiquitin ligases play pivotal roles in the regulation of NLR family pyrin domain containing 3 (NLRP3) inflammasome activation, a critical process in innate immunity and inflammatory responses.
