The study identifies TRIM15 as a key driver in the development of obesity‐associated esophageal adenocarcinoma (EAC). Mechanistically, TRIM15 degrades YY2 through the proteasome pathway, suppressing FOXRED1 transcription and ultimately accelerating tumor proliferation.

BRCA1 Is a Histone-H2A-Specific Ubiquitin Ligase
The RING domain proteins BRCA1 and BARD1 comprise a heterodimeric ubiquitin (E3) ligase that is required for the accumulation of ubiquitin conjugates at sites of DNA damage and for silencing at DNA satellite repeat regions. Despite its links to chromatin,

This study reveals that circPVT1 promotes the renal cell carcinoma lung metastasis and tumor progression through encoding the cP104aa peptide and directly binds with EIF4A3, which jointly regulates the target gene c‐MYC. Abstract Circular RNA (circRNA) plays a pivotal role in the pathogenesis of renal cell carcinoma (RCC).

Calhm6 drives M2 macrophage polarization via the Chp1‐Camk4‐Creb1 axis, suppressing inflammation through calcium‐dependent ectosomal delivery. Calhm6 deficiency enhances M1 responses, boosting bactericidal activity but exacerbating tissue damage. LPS/IFNγ upregulate Calhm6 via Irf1, while IL‐4/Stat6 inhibits it, balancing immune outcomes.

RNF8/UBC13 ubiquitin signaling suppresses synapse formation in the mammalian brain
Ubiquitin ligases play critical roles in neuronal connectivity in the brain. Here, Valnegri and colleagues show that ubiquitin ligase RNF8 and ubiquitin-conjugating enzyme UBC13 regulate synapse number in cerebellar granule neurons and rodent cerebellar ...

A Tyrosine Switch on NEDD4-2 E3 Ligase Transmits GPCR Inflammatory Signaling
Summary: Ubiquitination is essential for protein degradation and signaling and pivotal to many physiological processes. Ubiquitination of a subset of G-protein-coupled receptors (GPCRs) by the E3 ligase NEDD4-2 is required for p38 activation, but how ...

The E3 ubiquitin ligase c-IAP1 regulates PCSK9-mediated LDLR degradation: Linking the TNF-α pathway to cholesterol uptake [PDF]
Proprotein convertase subtilisin/kexin type 9 (PCSK9), in addition to LDLR (low-density lipoprotein receptor) and APOB (apolipoprotein B), is one of three loci implicated in autosomal dominant hypercholesterolaemia (ADH)^1^.

Mobile messenger RNAs (mRNAs) can travel long distances, serving as systemic signals that participate in plant growth and stress acclimation. This work determines that PHR proteins mediate the Pi starvation response (PSR)‐specific long‐distance transport of mRNAs and that these PSR‐specific mobile mRNAs represent noncoding transcripts in recipient ...

Antagonistic roles of ubiquitin ligase HEI10 and SUMO ligase RNF212 regulate meiotic recombination
Crossover recombination facilitates the accurate segregation of homologous chromosomes during meiosis. In mammals, poorly characterized regulatory processes ensure that every pair of chromosomes obtains at least one crossover, even though most ...
