Isolation of Extracellular Vesicles Derived from Mesenchymal Stromal Cells by Ultracentrifugation

open access: yesBio-Protocol, 2020
Extracellular vesicles (EVs) are a heterogeneous group of membranous vesicles that differ on their biogenesis and release pathways, such as exosomes, microvesicles and apoptotic bodies.

Isolation, characterisation and experimental evolution of phage that infect the horse chestnut tree pathogen, Pseudomonas syringae pv. aesculi [PDF]

open access: yes, 2020
Bleeding canker of horse chestnut trees is a bacterial disease, caused by the bacterium Pseudomonas syringae pv. aesculi, estimated to be present in ~ 50% of UK horse chestnut trees.

Autophagosome marker, LC3, is released extracellularly via several distinct pathways

open access: yesFEBS Open Bio, EarlyView.
This study establishes a novel HiBiT‐tagging system for ultrasensitive detection of LC3, revealing multiple pathways for its extracellular secretion. It demonstrates that LC3 is released via both autophagy‐dependent and ‐independent mechanisms, including a novel route for nonlipidated LC3‐I.

Practical Aspects of Multiwavelength Analytical Ultracentrifugation

open access: yesInstruments, 2019
Open-source Multiwavelength Analytical Ultracentrifugation (MWL-AUC) detection systems have been evolving for over a decade. Continual advances emerging out of several research groups have brought the instrumentation technology to increasingly higher ...

RNA‐Micelles as Self‐Assembling Structures for Efficient Co‐Delivery of Synergistic siRNA and Nucleoside Analogues to Treat CRC Lung Metastasis

open access: yesAdvanced Functional Materials, EarlyView.
Two kinds of self‐assembled RNA micelles were used to co‐deliver synergistic siRNA and nucleoside analogues for the treatment of colorectal cancer lung metastases. Near‐complete elimination of lung cancer metastases was confirmed in an orthotopic lung metastasis model constructed using human colorectal cancer lung metastases patient surgical samples to

A novel three step protocol to isolate extracellular vesicles from plasma or cell culture medium with both high yield and purity

open access: yesJournal of Extracellular Vesicles, 2020
Extracellular vesicles (EV) are membrane encapsulated nanoparticles that can function in intercellular communication, and their presence in biofluids can be indicative for (patho)physiological conditions.

Alternative Method for HDL and Exosome Isolation with Small Serum Volumes and Their Characterizations

open access: yesSeparations, 2021
High-density lipoprotein (HDL) and exosomes are promising sources of biomarkers. However, the limited sample volume and access to the ultracentrifuge equipment are still an issue during HDL and exosome isolation.

Cytoplasmic DNA in the unfertilized sea urchin egg: Physical properties of circular mitochondrial DNA and the occurrence of catenated forms [PDF]

open access: yes, 1968
The mitochondrial DNA in the unfertilized egg of the sea urchin Lytechinus pictus is present in an amount approximately seven times that of the haploid nuclear DNA.(1) The mitochondrial DNA has a higher buoyant density than the nuclear DNA and consists ...

High‐Resolution Characterization of Protein‐Conjugated, mRNA‐Loaded Lipid Nanoparticles by Analytical Ultracentrifugation

open access: yesAdvanced Functional Materials, EarlyView.
Lipid nanoparticles containing messenger RNA are characterized by sedimentation velocity analytical ultracentrifugation using UltraScan's new Custom Grid algorithm to provide multi‐dimensional distributions for partial specific volume (particle density), molar mass, sedimentation, diffusion, and hydrodynamic radius.

Particle size and protein content of six fractions of the Sf > 20 plasma lipoproteins isolated by density gradient centrifugation

open access: yesJournal of Lipid Research, 1969
A procedure is described for the separation of plasma Sf > 400 and Sf 20-400 lipoproteins each into three fractions. Serum samples are overlayered with a sodium chloride density gradient in a preparative ultracentrifuge tube and thin layers are removed ...
