Modulation of Homer1 EVH1 domain internal dynamics by putative autism‐associated mutations

open access: yesFEBS Letters, EarlyView.
The putative autism‐associated M65I and S97L variants of the EVH1 domain of the postsynaptic scaffold protein Homer1 do not exhibit substantial changes in their overall structure or partner binding. Both of them, but especially the M65I variant, show altered internal dynamics relative to the wild‐type domain on the μs‐ms timescale, indicated by the ...

TRAIL‐PEG‐Apt‐PLGA nanosystem as an aptamer‐targeted drug delivery system potential for triple‐negative breast cancer therapy using in vivo mouse model

open access: yesMolecular Oncology, EarlyView.
Aptamers are used both therapeutically and as targeting agents in cancer treatment. We developed an aptamer‐targeted PLGA–TRAIL nanosystem that exhibited superior therapeutic efficacy in NOD/SCID breast cancer models. This nanosystem represents a novel biotechnological drug candidate for suppressing resistance development in breast cancer.

Boundary Value Problem For the Equation of Mixed Type with Singular Coefficient in the Domain where the Elliptical Part Is a Half-band

open access: yesVestnik Samarskogo Gosudarstvennogo Tehničeskogo Universiteta. Seriâ: Fiziko-Matematičeskie Nauki, 2009
Boundary value problem is studied in the unbounded domain. Uniqueness of the problem is proven with the help of the extreme principle. Existence of the problem is established by methods of separation of variables and integral equations.

ON ELLIPTIC PROBLEMS IN DOMAINS WITH UNBOUNDED BOUNDARY [PDF]

open access: yesProceedings of the Edinburgh Mathematical Society, 2006
AbstractThe paper deals with problems of the type $-\Delta u+a(x)u=|u|^{p-2}u$, $u\gt0$, with zero Dirichlet boundary condition on unbounded domains in $\mathbb{R}^N$, $N\geq2$, with $a(x)\geq c\gt0$, $p\gt2$ and $p\lt2N/(N-2)$ if $N\geq3$. The lack of compactness in the problem, related to the unboundedness of the domain, is analysed. Moreover, if the

Generalized Active Information: Extensions to Unbounded Domains [PDF]

open access: yesBIO-Complexity, 2020
In the last three decades, several measures of complexity have been proposed. Up to this point, most of such measures have only been developed for finite spaces. In these scenarios the baseline distribution is uniform. This makes sense because, among other things, the uniform distribution is the measure of maximum entropy over the relevant space ...

Subtype‐specific enhancer RNAs define transcriptional regulators and prognosis in breast cancers

open access: yesMolecular Oncology, EarlyView.
This study employed machine learning methodologies to perform the subtype‐specific classification of RNA‐seq data sets, which are mapped on enhancers from TCGA‐derived breast cancer patients. Their integration with gene expression (referred to as ProxCReAM eRNAs) and chromatin accessibility profiles has the potential to identify lineage‐specific and ...

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

The Existence and Uniqueness of an Entropy Solution to Unilateral Orlicz Anisotropic Equations in an Unbounded Domain

open access: yesAxioms, 2020
The purpose of this work is to prove the existence and uniqueness of a class of nonlinear unilateral elliptic problem (P) in an arbitrary domain, managed by a low-order term and non-polynomial growth described by an N-uplet of N-function satisfying the ...

Special wave finite and infinite elements for the solution of the Helmholtz equation [PDF]

open access: yes, 2003
The theory and the formulation of the special wave finite elements are discussed, and the special integration schemes for the elements are developed.

A novel quinazolinone insulin receptor inhibitor and its synergy with an EGFR inhibitor in glucose‐driven glioblastoma

open access: yesMolecular Oncology, EarlyView.
The novel styrylquinazolinone‐based molecule W1B effectively suppresses glioblastoma by inhibiting IGF1R and EGFR. In high‐glucose microenvironments driving tumor resistance, W1B acts synergistically with the EGFR inhibitor dacomitinib. This combination safely blocks compensatory survival signaling in zebrafish xenograft models. Showcasing promising in
