Discerning protein pools by selective staining with self‐labeling tags
Cell surface proteins have an intra‐ and extracellular pool. Combining genetic fusion to self‐labeling tags that can be addressed with small molecule fluorophores allows separating these pools. We highlight recent developments and techniques for state‐of‐the‐art interrogation of cell surface proteins in the complex tissue setting.

Calibration methods for a dual-wavelength interferometer system
Multiple wavelength interferometry has long been considered an option for the measurement of large aspheric slope departures. In particular, a synthetic wavelength offers a variety of approaches by which large phase excursions can be unwrapped.

Raman‐based label‐free microscopic analysis of the pancreas in living zebrafish larvae
Forward stimulated Raman scattering (F‐SRS) and epi coherent anti‐Stokes Raman scattering (E‐CARS) allow label‐free discrimination of distinct subcellular structures in the pancreas of living zebrafish larvae. Given the straightforward applicability, we anticipate broad implementation of Raman microscopy in other organs and across various biomedical ...

Time‐resolved X‐ray solution scattering captures how proteins change shape in real time under near‐native conditions. This article presents a practical workflow for light‐triggered TR‐XSS experiments, from data collection to structural refinement. Using a calcium‐transporting membrane protein as an example, the approach can be broadly applied to study ...

Due to the strong absorption of water vapor at wavelengths of 1350–1420 nm and 1820–1940 nm, under normal atmospheric conditions, the actual digital number (DN) response curve of a hyperspectral imager deviates from the Gaussian shape, which ...

This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Calibration-free wavelength modulation spectroscopy based on even-order harmonics
This paper proposes a novel and rapid calibration-free wavelength modulation spectroscopy algorithm based on even-order harmonics. The proposed algorithm, analytically deduced from Voigt line-shape function, only involves simple algebraic operations to ...
