Calpain small subunit homodimerization is robust and calcium‐independent

open access: yesFEBS Letters, EarlyView.
Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Learning max-weight discriminative forests [PDF]

open access: yes2008 IEEE International Conference on Acoustics, Speech and Signal Processing, 2008
We present a method for sequential learning of increasingly complex graphical models for discriminating between two hypotheses. We generate forests for each hypothesis, each with no more edges than a spanning tree, which optimize an information-theoretic criteria.

Weight-Based Health Care Discrimination and Cervical Cancer Screening Among Black Sexual and Gender Minoritized Assigned Female at Birth Adults in the United States

open access: yesHealth Equity
Introduction: Black sexual and gender minoritized (SGM) people assigned female at birth (AFAB) face notable barriers to cervical cancer screening, including racism, heterosexism, and cisgenderism.

Missing the target: including perspectives of women with overweight and obesity to inform stigma‐reduction strategies

open access: yesObesity Science & Practice, 2017
Summary Objective Pervasive weight stigma and discrimination have led to ongoing calls for efforts to reduce this bias. Despite increasing research on stigma‐reduction strategies, perspectives of individuals who have experienced weight stigma have rarely

Reconstructing enzyme evolution by protein engineering

open access: yesFEBS Letters, EarlyView.
Natural enzyme evolution can be retraced by protein engineering methods such as directed evolution, rational design, and ancestral sequence reconstruction. These approaches reveal how enzymes emerged from ligand‐binding scaffolds, developed varying substrate preferences, formed oligomeric complexes, adapted to environmental changes, and evolved novel ...

Evidence-based recommendations to assist adults with depression to become lifelong movers

open access: yesHealth Promotion and Chronic Disease Prevention in Canada, 2020
IntroductionWeight-related social stigma is associated with adverse health outcomes. Health care systems are not exempt of weight stigma, which includes stereotyping, prejudice and discrimination.

Longitudinal circulating tumor DNA profiling in patients with advanced endometrial cancer using an off‐the‐shelf targeted NGS panel

open access: yesMolecular Oncology, EarlyView.
Intratumour heterogeneity complicates precision management of advanced endometrial cancer. Circulating tumor DNA (ctDNA) offers a minimally invasive strategy to capture tumor evolution and therapeutic resistance. Here, we compare tumor‐agnostic NGS with tumor‐informed ddPCR, outlining their relative sensitivity, concordance, and clinical implications ...

Obesity‐Related Beliefs, Concerns, and Stigmatizing Perceptions Among Adults Living With Obesity

open access: yesObesity Science & Practice
Background Weight stigma represents a barrier to effective obesity management and to achieving health equity. Although most research has focused on high‐income countries, data from Latin America remain scarce.

Weightism in Asia: A Narrative Review and Implications for Practice

open access: yesJournal of Obesity & Metabolic Syndrome
Weightism, also known as weight-related discrimination, is pervasive and believed to be one of the socially accepted types of discrimination in Asia.

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...
