Electrochemical and theoretical evaluation of the interaction between dna and amodiaquine: evidence of the guanine adduct formation

open access: yesQuímica Nova, 2010
The electrochemical behavior of the interaction of amodiaquine with DNA on a carbon paste electrode was studied using voltametric techniques. In an acid medium, an electroactive adduct is formed when amodiaquine interacts with DNA.

Is amodiaquine failing in Rwanda? Efficacy of amodiaquine alone and combined with artesunate in children with uncomplicated malaria [PDF]

open access: yesTropical Medicine & International Health, 2004
SummaryWe investigated the safety and efficacy of amodiaquine alone (AQ) and combined with artesunate (AQ + AS) in 308 Rwandan children 6–59 months old with uncomplicated Plasmodium falciparum malaria attending three sentinel sites. The two treatment regimes were well tolerated and no serious adverse events were recorded. After excluding new infections,

Artemisinin-based combination therapy for uncomplicated Plasmodium falciparum malaria in Colombia

open access: yesMalaria Journal, 2007
Background Artemisinin-based combination therapy (ACT) is being widely promoted as a strategy to counteract the increase in Plasmodium falciparum antimalarial drug resistance.

Field-adapted sampling of whole blood to determine the levels of amodiaquine and its metabolite in children with uncomplicated malaria treated with amodiaquine plus artesunate combination

open access: yesMalaria Journal, 2009
Background Artemisinin combination therapy (ACT) has been widely adopted as first-line treatment for uncomplicated falciparum malaria. In Uganda, amodiaquine plus artesunate (AQ+AS), is the alternative first-line regimen to Coartem® (artemether ...

Amodiaquine-induced Agranulocytosis: Toxic Effect of Amodiaquine in Bone Marrow Cultures In Vitro [PDF]

open access: yesBMJ, 1973
A case of agranulocytosis is reported in which amodiaquine, to which the patient had been exposed, was found to be toxic to the patient's bone marrow cells when these were cultured in an agar colony system in vitro. This technique should be investigated in other patients with agranulocytosis as a possible means of detecting toxic agents.

Interactions between DNA purinic bases and amodiaquine: A theoretical approach

open access: yesOrbital: The Electronic Journal of Chemistry, 2010
We study theoretically the amodiaquine-adenine and amodiaquine-guanine adducts formation using Density Functional Theory (B3LYP) and the 6-31G(d) basis set for the geometry optimizations and 6-31+G(d,p) for the analysis of the global indexes ...

Glutathione S-Transferase P1 Protects Against Amodiaquine Quinoneimines-Induced Cytotoxicity but Does Not Prevent Activation of Endoplasmic Reticulum Stress in HepG2 Cells

open access: yesFrontiers in Pharmacology, 2018
Formation of the reactive amodiaquine quinoneimine (AQ-QI) and N-desethylamodiaquine quinoneimine (DEAQ-QI) plays an important role in the toxicity of the anti-malaria drug amodiaquine (AQ).

INTRAVENOUS AMODIAQUINE AND ORAL AMODIAQUINE/ERYTHROMYCIN IN THE TREATMENT OF CHLOROQUINE-RESISTANT FALCIPARUM MALARIA

open access: yesThe Lancet, 1985
In eastern Thailand, 14 adults with moderately severe falciparum malaria were treated with intravenous amodiaquine dihydrochloride, loading dose 10 mg base/kg infused over 4 h followed by three further intravenous infusions of 5 mg base/kg at 24, 48, and 72 h.

Comparative efficacy of artesunate and sulphadoxine-pyrimethamine combination with artesunate and amodiaquine combination in uncomplicated falciparum malaria in children

open access: yesPaediatrica Indonesiana, 2008
Background Malaria is still an important cause of mortality and morbidity in children and adults in tropical countries. Multidrug resistance againts chloroquine and sulphadox- ine-pyrimethamine had brought to an introduction of artemisinin-based ...
