TRAIL分子胞外区基因工程蛋白的制备与鉴定

目的 制备人TRAIL配体胞外区基因工程蛋白。方法 应用RT PCR技术从激活的人外周血淋巴细胞总RNA中扩增TRAIL配体胞外区cDNA ,克隆入PCR2 .1 载体 ,测序验证后用基因重组法构建了TRAIL配体胞外区和组氨酸盒的原核表达质粒pQE hTRAIL。将重组质粒转入大肠杆菌M15中表达 ,经 1mmol LIPTG在 37℃条件下诱导 4h ,亲和层析柱纯化。用SDS -PAGE电泳和Westernblot鉴定表达产物。结果 所表达融合蛋白为人TRAIL配体胞外区分子 ,分子量 30kD ...

ABSTRACT SnTe is considered a potential alternative to PbTe for thermoelectric applications. However, excessive carrier concentration and thermal conductivity result in poor thermoelectric performance. Herein, p‐type SnTe‐based materials Sn0.8‐xMn0.1Pb0.1SbxTe were synthesized, and the combined effects of Sb, Mn, and Pb were systematically investigated.

Properties of double-liquid phase fermentation system for extracellular polysaccharide production of Ganoderma lucidum(灵芝胞外多糖双液相发酵体系的特性)

用豆油作为氧载体加入发酵液中形成双液相发酵体系,以改善灵芝胞外多糖的发酵状况.实验表明:常见的氧载体材料在双液相发酵过程中不为灵芝菌体所利用,仅起到促进溶氧的效果;在灵芝胞外多糖的双液相发酵体系中, KLa值明显提高,溶氧水平(DO)有显著改善,发酵液的运动黏度µ有所降低,有助于降低搅拌能耗;氧载体改善了质量传递,提高了细胞的代谢水平,使胞外多糖的产量提高了44%,发酵周期缩短了约17%.说明双液相发酵体系对灵芝胞外多糖发酵是一种新型有效的途径.
