外泌体microRNA-212/132与创伤性颅脑损伤严重程度及Rotterdam CT评分的相关性研究

目的 探讨创伤性颅脑损伤(TBI)患者外泌体microRNA-212/132(miR-212、miR-132)表达水平,及其与患者脑损伤严重程度和Rotterdam CT评分的相关性。方法 76例TBI患者根据格拉斯哥昏迷评分(GCS)分为轻度组(13-15分)19例,中度组(9-12分)35例,重度组(3-8分)22例。另选择38例体检健康者作为对照。所有受试者均给予颅脑CT检查并行Rotterdam CT评分;提取血浆外泌体RNA并采用实时荧光定量PCR技术检测miR-212、miR-132表达水平,

基于二代测序的RhD阳性个体RHD基因mRNA选择性剪接分析

目的本研究旨在对RhD阳性个体中红细胞特异性表达的RHD基因不同转录本进行分析,并对选择性剪接发生的机制进行初步探讨。方法利用体外有核红细胞培养体系分离并培养RhD阳性个体的有核红细胞,使用逆转录聚合酶链反应(RT-PCR)技术对RhD阳性个体RHD基因外显子6至3’非编码区扩增后进行二代测序,分析mRNA转录本,并采用生物信息学方法分析RHD基因所有外显子5’及3’剪接位点的最大熵值。结果RHD基因的转录本以正常全长mRNA为主,除此之外还存在其他8种异常转录本。按照表达量从多到少,分别为外显子7缺失、

Lactacystin对体外C6胶质瘤细胞超微结构影响的研究

目的探讨蛋白酶体抑制剂Lactacystin对体外C6胶质瘤细胞超微结构的影响。方法Lactacystin处理体外培养的C6胶质瘤细胞,HE染色后光镜观察细胞的死亡情况,锇铀铅染色后透射电镜观察细胞超微结构变化。结果Lactacystin处理体外C6胶质瘤细胞24 h后,光镜下见胞浆浓缩深染,胞核染色质致密浓缩,透射电镜下见细胞表面微绒毛消失,核皱缩,染色质致密浓缩、边集。结论蛋白酶体抑制剂Lactacystin可以诱导体外C6胶质瘤细胞凋亡。
