应用聚合酶链反应技术扩增血痕DNA,根据扩增产物中有无aPoB基因的数量可变重复序列(VNTR)及其扩增片段的大小即可同时确定其种属特异性和DNA的多态性。实验结果显示人血痕可见扩增带并具有多态性,而猴猪狗猫鸡血痕无扩增带。

从1~2cm人发干中提取基因组DNA,以募核苷酸ya、y4_为引物,经过聚合酶链反应,扩增Y染色体特异的片段(446bP)。根据有无该片段的存在来确定发干的性别。这种方法取材方便,灵敏度高、易于推广。本文还报道了科用本方法检测某案例中5根发干的性别,为侦破工作提供了线索。

采用聚合酶链反应(PCR)结合聚丙烯酰胺凝胶电泳(PAGE),检测PCR产物异源双链形成情况,诊断和产前诊断B珠蛋白生成障碍性贫血(β地中海贫血)CD41-42(-CTTT)突变。此方法简便快速,可区分有无突变,并能鉴别纯合子与杂合子。应用本方法进行了4例产前诊断。对此方法的优点和局限性及应用前景进行了讨论。
