【目的】探索一种利用随机引物聚合酶链反应指纹法技术进行变形链球菌基因分型的方法。【方法】利用变形链 球菌不同血清型标准株和通用引物5′-TGCCGAGCTG-3′, 研究了在不同条件下进行聚合酶链反应的结果, 寻找满意的聚合酶 链反应条件。【结果】找到了较为满意的聚合酶链反应条件:94 ℃ 30 s , 36 ℃ 30 s, 72 ℃ 1 min, 反应进行45 个循环。【结论】 随机引物聚合酶链反应指纹法技术在一定的控制条件下可用于变形链球菌的鉴定和分型。

聚合酶链反应及染色法在口腔念珠菌白斑诊断中的应用 [PDF]
【目的】探讨聚合酶链反应技术在检测口腔白斑中念珠菌感染的应用。【方法】应用组织化学染色法和聚合酶链 反应技术对口腔白斑是否存在念珠菌进行检测。【结果】107 例口腔白斑, 经PAS 和六胺银染色, 16.8%(18/ 107)的口腔白斑 检出念珠菌菌丝。念珠菌细胞色素P450基因片段PCR 检出率为10 %(11/ 107), 其中的白色念珠菌EO3 基因片段的PCR 检出 率为6.5%(7/ 107)。【结论】组织化学染色法能直观显示口腔白斑中念珠菌形态分布特点以及所引起的组织学改变, 而PCR

ABSTRACT Objective Placental infection with severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) can lead to placental insufficiency and in‐utero fetal death (IUFD). The objective of this study was to confirm and quantify the extent to which fetoplacental infection with SARS‐CoV‐2 is a cause of fetal death.

Amplification of Ovis aries papillomavirus type 2 DNA from an ovine cutaneous fibropapilloma
Seven of 60 Perendale sheep within a flock developed single or multiple exophytic masses on their distal hind limbs. A mass was excised from one sheep and histological evaluation revealed epidermal and mesenchymal proliferation, papillomavirus‐induced keratinocyte changes and marked keratohyalin clumping.

【目的】构建人瘦素基因 cDNA 克隆, 并进行序列分析。【方法】用逆转录聚合酶链反应扩增人瘦素基因 cDNA , 获得片段( 640 bp)连接至 pUC19 载体, 并转化大肠杆菌 DH5α。以末端终止法进行序列测定。【结果】所克隆的人瘦素 cDNA 含全长的人瘦素基因编码区,仅 287 位的腺苷酸被鸟苷酸代替, 导致 96 位编码谷氨酰胺的密码子 CAA 改变为编码精氨酸的 CGA, 其意义尚待阐明。【结论】以逆转录聚合酶链反应方法成功地构建了人瘦素基因 cDNA 克隆。
