目的明确血管性认知损伤大鼠脑内SIRT1表达及氧化应激指标SOD及MDA的变化情况。方法 2-VO法建立慢性脑缺血大鼠模型,随机分为假手术组和慢性脑缺血组,于术后6周、9周、12周,利用Morris水迷宫试验进行认知评估,实时定量PCR、免疫组化及Western blot检测大鼠脑内SIRT1表达,同时进行SOD、MDA水平测定。结果 2-VO术后6周、9周和12周,慢性脑缺血大鼠SIRT1蛋白表达明显下降,以缺血6周时最明显;慢性脑缺血时大鼠脑内SOD降低而MDA升高,显著差异仅发生在脑缺血6周 ...
