3-Mercaptopyruvate Sulfurtransferase Produces Hydrogen Sulfide and Bound Sulfane Sulfur in the Brain
Antioxidants & Redox Signaling, 2009Hydrogen sulfide (H(2)S) is a synaptic modulator as well as a neuroprotectant. Currently, pyridoxal-5'-phosphate (PLP)-dependent cystathionine beta-synthase (CBS) is thought to be the major H(2)S-producing enzyme in the brain. We recently found that brain homogenates of CBS-knockout mice, even in the absence of PLP, produce H(2)S at levels similar to ...

[38] 3-Mercaptopyruvate sulfurtransferase
1981Publisher Summary 3-Mercaptopyruvate sulfurtransferase catalyzes the cleavage of a carbon–sulfur bond and the transfer of the sulfur atom from 3-mercaptopyruvate to any of a variety of thiophiles, including thiols, cyanide, sulfite, and sulfinates. This chapter deals with the purification of the 3-mercaptopyruvate sulfurtransferase from the bovine ...

A steady-state kinetic analysis of 3-mercaptopyruvate sulfurtransferase (EC 2.8.1.2) using cyanide as the sulfur-acceptor substrate was performed. Measurement of pyruvate production gave initial velocity patterns and secondary plots characteristic of a rapid equilibrium-ordered sequential mechanism.

3-mercaptopyruvate sulfurtransferase (EC 2.8.1.2): A simple assay adapted to human blood cells
Clinica Chimica Acta, 1974Abstract A product of cysteine catabolism, 3-mercaptopyruvate, is enzymatically degraded to sulfur and pyruvate by a sulfurtransferase (EC 2.8.1.2.) present in human red and white blood cells. A simple sulfurtransferase assay is reported which takes advantage of the fact that pyruvate is conveniently measured enzymatically by lactate dehydrogenase ...

Sulfhydration of reactive cysteines in target proteins is now recognized as a major route by which H2S mediates signal transduction and regulates various cellular processes. Among the enzymatic systems permitting the formation of cysteine persulfide from nonactivated sulfur compounds, 3-mercaptopyruvate sulfurtransferases can be considered as a model ...

The present experiments were carried out to investigate the expression of 3-mercaptopyruvate sulfurtransferase (3MST) in medulla oblongata of rats and effects of chronic intermittent hypoxia (CIH) on its expression. Sprague Dawley adult rats were randomly divided into two groups, including control (Con) group and CIH group. The endogenous production of

H2S (hydrogen sulfide) has recently been recognized as a signalling molecule as well as a cytoprotectant. We recently demonstrated that 3MST (3-mercaptopyruvate sulfurtransferase) produces H2S from 3MP (3-mercaptopyruvate). Although a reducing substance is required for an intermediate persulfide at the active site of 3MST to release H2S, the substance ...

3-Mercaptopyruvate sulfurtransferase activity in guinea pig and rat tissues.
Physiological chemistry and physics and medical NMR, 1985Activities of 3-mercaptopyruvate sulfurtransferase (EC 2.8.1.2) in guinea pig tissues were determined and compared with those in the corresponding rat tissues. The activities in guinea pig tissues were found to be very low. The activity in the liver was 145.9 mumol pyruvate formed per g of fresh tissue per 15 min.

Inhibition of 3-mercaptopyruvate sulfurtransferase enhances CD8⁺ T-cell antitumor immunity
Abstract Hydrogen sulfide (H₂S) is a redox-active gasotransmitter implicated in tumor progression and immune regulation. The enzyme 3-mercaptopyruvate sulfurtransferase (3-MST) is a key contributor to endogenous H₂S and polysulfide production, but its role in tumor–immune interactions remains poorly defined.Muriel Urwyler +13 more
