Nine healthy subjects (7 men; 2 women) received single 20-mg IV injections of 4-aminopyridine (4-AP). Six of the subjects received the same dose in the form of enteric-coated tablets and four the same dose in uncoated tablets; treatments were at least 2 wk apart.

INDO theoretical studies of 4-aminopyridine and protonated 4-aminopyridine
Journal of the Chemical Society, Perkin Transactions 2, 1975Geometry optimized INDO calculations have been performed on 4-aminopyridine (2) and protonated 4-aminopyridine (1) in order to compare their optimized geometries, π-bond orders, C(4)–NH2 rotational barriers, charge densities, and π-electron distributions. It had previously been shown that 4-aminopyridine falls far off a linear correlation of ΔG° and ΔS°

4‐Aminopyridine in multiple sclerosis
Neurology, 1991In an earlier study, we demonstrated efficacy of single oral doses of 4-aminopyridine (4-AP) in improving motor and visual signs in multiple sclerosis (MS) patients for a mean of 4.97 hours. We attempted to determine whether efficacy could safely be prolonged using multiple daily doses over several days by administering 7.5 to 52.5 mg 4-AP to 17 ...

Characterization of 4-Aminopyridine in Overdose
Journal of Toxicology: Clinical Toxicology, 1994We report three cases of 4-aminopyridine overdose resulting in seizure activity. All patients were on therapeutic regimens for the treatment of multiple sclerosis. All patients were responsive to intensive supportive care, although the length of toxicity was prolonged more than 24 hours in one patient.

Pharmacokinetics of 4-aminopyridine in horses
American Journal of Veterinary Research, 1984SUMMARY The pharmacokinetics of 4-aminopyridine (4-ap), a drug capable of antagonizing nondepolarizing neuromuscular blocking drugs, as well as several classes of injectable sedative and anesthetic agents, were studied in 6 intact, awake horses. Plasma samples were assayed for 4-ap over a frequent sampling schedule for 8 hours after iv administration ...

Long-term potentiation and 4-aminopyridine
Cellular and Molecular Neurobiology, 1985Long-term potentiation (LTP) of excitatory postsynaptic potentials (epsp's) was investigated with extracellular field potential recording in hippocampal slices from rats. In the presence of 100 microM 4-aminopyridine (4-AP) the probability of eliciting LTP was unchanged or increased; the extent of potentiation was not significantly different from ...

Pharmacokinetics of 4-aminopyridine in cattle
American Journal of Veterinary Research, 1984SUMMARY Concentrations of 4-aminopyridine hydrochloride in plasma of cattle were measured over an 8-hour period following a bolus IV injection (0.3 mg/kg). The drug was assayed by a gas-liquid chromatographic method using a nitrogen-phosphorus detector.

Interaction of antidepressants with 4-aminopyridine
Life Sciences, 1992Systemic administration of 4-Aminopyridine at a dose of 4 mg/kg (4-AP) induces hypothermia in mice. Scopolamine (ED50 = 0.26 mg/kg) and two tricyclic antidepressants, desipramine (ED50 = 1.82 mg/kg) and IM/P/3/4 (ED50 = 8.95 mg/kg) completely antagonize 4-AP-induced hypothermia, whereas minaprine (0.1-0.25 mg/kg), a non-tricyclic antidepressant ...
