The putative 5-HT agonist 8-hydroxy-2-(di-n-propylamino)tetralin (8-OHDPAT) produced in rats an increase in the number of shocks accepted in a modified Vogel's conflict test design. Subchronic pretreatment with p-chlorophenylalanine (PCPA) similarly caused release of the punished behavior.

The effects of intra-amygdala injection of midazolam (20 nmol) and 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT; 8 and 16 nmol) were investigated in rats submitted to the elevated T-maze, a new animal model of anxiety. This test allows the measurement, in the same rat, of conditioned and unconditioned fear/anxiety responses.

8-OH- DPAT (8-hydroxy-2-[di-n-propylamino] tetralin) is a novel aminotetralin derivative which has been proposed to be a serotonin (5-HT) agonist devoid of dopamine agonist effects. We now report that the administration of 8-OH- DPAT , like known 5-HT agonists, produced a rapid elevation of serum prolactin concentrations in male rats.

Racemic 8-OH-DPAT, (R,S)-8-hydroxy-2-(di-n-propylamino)tetralin, has become the prototype 5-HT1A receptor agonist. The enantiomers of 8-OH-DPAT have similar affinities to the 5-HT1A receptor, but the (R)-enantiomer is a full agonist, whereas the (S)-enantiomer is a partial agonist.

The effects of the 5-HT1A receptor agonist, 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT) on the behaviour of mice were studied. 8-OH-DPAT given i.v. in doses greater than 1 mg/kg induced the distinct 5-HT syndrome, including head weaving, hindlimb abduction, forepaw treading and tremor. Abstract A new synthesis method for 8-OH-DPAT, a specific agonist at the 5-HT1A serotonin receptor, is described. It employs the Curtius degradation of a tetralin carboxylic acid easily prepared from (2-methoxy benzyl) succinic acid by Friedel-Crafts cyclisation.
