Acid Phosphatase in Eyes with Pseudoexfoliation

We studied the activity of acid phosphatase in the anterior segment of the eye with pseudoexfoliation histochemically and biochemically. Light and electron microscopic histochemistry revealed lead precipitates indicating acid phosphatase activity in pseudoexfoliative materials.

Abstract Extracts of the alga Enteromorpha linza hydrolysed glucose-6-phosphate, p -nitrophenylphosphate 2′-, 3′-, and 5′-adenosine monophosphates with an optimum at pH 5. Cytidine and uridine-5′-nucleoside diphosphates, and 2′-, and 3′-adenosine monophosphates were relatively poorly hydrolysed.

GOMORI1and Takamatsu2in 1939 showed that the presence of phosphatase can be demonstrated in tissue sections. Their work referred to alkaline phosphatase; however, later, Gomori3described a method by which acid phosphatase, too, can be made visible in sections. Wolf, Kabat and Newman4examined the nerve tissue for acid phosphatase.

Acid phosphatases are ubiquitous enzymes present in almost all tissues. Isoenzyme 2 and the antigenically related but deglycosy-lated isoenzyme 4 are found predominantly in prostatic tissue. The association of elevated acid phosphatase with carcinoma of prostate is of long standing but the specificity of kinetic enzyme assays has been less than perfect.

Abstract Two acid phosphatases of human erythrocytes have been detected and separated by means of gel filtration on polymerized dextrans. The two enzymes, given the trivial names E 1 phosphatase and E 2 phosphatase, differ in molecular weight, pH optima, metal requirements, and type of inhibitors.
