Ionomycin suppresses cancer cell growth by disrupting mitochondrial transcription

open access: yesFEBS Open Bio, EarlyView.
In this study, we identify a new function for the selective Ca2+ ionophore, ionomycin, as an inhibitor of mitochondrial transcription. Both total and nascent RNA analyses revealed that ionomycin treatment reduces the transcription of mitochondrial genes.

Down syndrome-associated leukaemias: current evidence and challenges

open access: yesTherapeutic Advances in Hematology
Children with Down syndrome (DS) are at increased risk of developing haematological malignancies, in particular acute megakaryoblastic leukaemia and acute lymphoblastic leukaemia.

Insights into molecular pathways for targeted therapeutics in acute leukaemia

open access: yesSwiss Medical Weekly, 2010
Despite the development of modern chemotherapeutic regimens, acute leukaemia remains incurable in the majority of adult patients and potential cure is associated with considerable side effects. Clinical and experimental research of the last two decades

GelMA‐based 3D spheroids recapitulate transcriptomic and functional hallmarks of myeloid sarcoma

open access: yesFEBS Open Bio, EarlyView.
GelMA 5% hydrogels support the formation of myeloid leukemia spheroids that recapitulate MS‐specific features, including G1 arrest, apoptosis, and ECM‐driven transcriptomic reprogramming. The 3D model mimicked soft‐tissue‐like stiffness and oxygen conditions, and transcriptomic convergence with primary MS samples confirmed its utility as a preclinical ...

Using cell‐free RNA to identify B‐ and T‐cell clonality for diagnosis and monitoring of B‐ and T‐cell neoplasms

open access: yesFEBS Open Bio, EarlyView.
Using peripheral blood for determining B‐cell or T‐cell clonality is more reliable when we use cell‐free RNA (cfRNA) because cells release blood significantly more RNA than DNA. Next‐generation sequencing (NGS) of cfRNA allows us to evaluate fragment cfRNA and evaluate clonality reliably without the need for prior determination of the specific dominant

The C‐terminal truncated splicing variant of NK1R negatively modulates substance P‐stimulated NK1R signaling

open access: yesFEBS Open Bio, EarlyView.
The neurokinin 1 receptor exists as full‐length (NK1L) and C‐terminally truncated (NK1S) splice variants. We show that NK1S heterodimerizes with NK1L, impairing Gαq coupling and Ca2+ mobilization while enhancing β‐arrestin1 recruitment. NK1S suppresses substance P‐driven gene expression and cell migration, revealing NK1S as an endogenous biased ...

Selective elimination of quiescent cancer cells by dequalinium chloride via ROS‐mediated ferroptosis

open access: yesFEBS Open Bio, EarlyView.
Dequalinium chloride (DQ) selectively eliminates quiescent cancer cells (QCCs) by amplifying mitochondrial ROS. QCCs exhibit elevated basal ROS and limited redox‐buffering capacity, making them vulnerable to further ROS increases. DQ‐induced ROS exceeds the tolerance threshold, triggering lipid peroxidation and ferroptosis, whereas proliferating cells ...

Leukaemia Incidence in Children and Adults in the Regions of Russia Most Highly Contaminated after the Chernobyl Nuclear Power Plant Accident [PDF]

open access: yes, 2003
BACKGROUND Leukaemia, especially the acute types predominant in children, may be caused by ionizing radiation. After the Chernobyl Nuclear Power Plant accident on 26 April 1986, parts of Belarus, Russia and the Ukraine were contaminated with ...

Rapidly progressing, fatal and acute promyelocytic leukaemia that initially manifested as a painful third molar: a case report

open access: yesJournal of Medical Case Reports, 2009
Introduction Acute promyelocytic leukaemia, an uncommon and devastating subtype of leukaemia, is highly prevalent in Latin American populations. The disease may be detected by a dentist since oral signs are often the initial manifestation.
