PEAR1 Promotes Glucose Metabolism Reprogramming in Sepsis‐Associated Acute Lung Injury via AARS1‐Mediated HIF‐1α Lactylation

open access: yesAdvanced Science, EarlyView.
This study revealed that a PEAR1/HIF‐1α/ glycolysis/lactate/H3K18la positive feedback loop in PMVECs that drives the development of S‐ALI. Mechanistically, PEAR1 mediates the binding of HIF‐1α to AARS1, leading to the lactylation of HIF‐1α, the primary lactylation site of which is K172.

Early application of awake extracorporeal membrane oxygenation in pneumocystis jirovecii pneumonia complicated with severe acute respiratory distress syndrome: a case report

open access: yesFrontiers in Medicine, 2023
IntroductionPatients suffering from severe acute respiratory distress syndrome (ARDS) are usually treated with mechanical ventilation. Extracorporeal membrane oxygenation (ECMO) has traditionally been considered a life-saving therapy and was reserved as ...

Chimeric IL‐6/4R‐LL37 Engineered Macrophages Achieve Synchronized Inflammation Control and Antimicrobial Defence in Sepsis

open access: yesAdvanced Science, EarlyView.
Engineered macrophages programmed in situ by LNP‐delivered IL‐6/4 fusion and LL37 mRNAs simultaneously dampen cytokine storm, promote M2‐like repair, and enhance direct bacterial killing in sepsis. This combinatorial strategy restores T cell and macrophage function, lowers organ bacterial burden, and improves survival, highlighting a precision, host ...

Informative Value of the Oxygenation Index in the Diagnosis of Acute Respiratory Distress Syndrome

open access: yesОбщая реаниматология, 2009
Objective: to study the informative value of the oxygenation index as a criterion for the diagnosis of acute respiratory distress syndrome (ARDS) of varying genesis. Subjects and methods. Seventy-two patients, including 27 women and 45 men, aged 19 to 65

MetaboSense: An Integrated Quantum Dot‐Mediated Aptamer Assay and Modular Microfluidic Platform for Continuous In‐Line Monitoring of Lung Metabolism During Ex Vivo Perfusion

open access: yesAdvanced Science, EarlyView.
MetaboSense is a first‐of‐its‐kind microfluidic platform enabling continuous, in‐line monitoring of glucose and lactate during ex vivo lung perfusion. Combining a quantum dot–mediated aptamer assay with mixing, depletion, and optofluidic detection modules, it achieves 60–s temporal resolution and shows strong agreement with standard blood gas analyzers,

Mass Spectrometry-based Proteomics in Acute Respiratory Distress Syndrome: A Powerful Modality for Pulmonary Precision Medicine

open access: yesChinese Medical Journal, 2016
Objective: Acute respiratory distress syndrome (ARDS) is an acute and lethal clinical syndrome that is characterized by hypoxemic respiratory failure and diffuse alveolar inflammatory damage. This review aimed to search and discuss the mass spectrometry (

A Biofabricated Human Acinus‐on‐a‐Chip Unveils Mechanotransductive Drivers of Ventilator‐Induced Lung Injury via Decoupling Volutrauma and Barotrauma

open access: yesAdvanced Science, EarlyView.
This bioinspired acinus‐on‐a‐chip recapitulates VILI pathology, revealing that volutrauma drives P53/NF‐κB pathways while barotrauma triggers mitochondrial‐Wnt dysregulation. A fibrotic transitional cell cluster was identified. Pharmacological interventions targeting these pathways significantly ameliorated injury, establishing a mechanobiological ...

Dual pKa Lipid Nanoparticles for Lung‐tropic mRNA Delivery and pH‐Programmed Endosomal Escape

open access: yesAdvanced Science, EarlyView.
Lipid nanoparticles’ bottleneck lies in low endosomal escape efficiency and liver‐dominant delivery after intravenous administration. To overcome these limitations, we present a stepwise pH‐programmed lipid nanoparticle system enabled by our newly synthesized ionizable lipid.

Targeting Neutrophils to Treat Acute Respiratory Distress Syndrome in Coronavirus Disease

open access: yesFrontiers in Pharmacology, 2020
This review describes targeting neutrophils as a potential therapeutic strategy for acute respiratory distress syndrome (ARDS) associated with coronavirus disease 2019 (COVID-19), a pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-
