A quantitative model‐based approach for adavosertib dose selection in patients with uterine serous carcinoma

British Journal of Clinical Pharmacology
Aims Adavosertib, a WEE1 inhibitor, has been studied for safety and efficacy as monotherapy and in combination therapy. The phase 2b ADAGIO trial (NCT04590248) evaluated adavosertib monotherapy for uterine serous carcinoma with a recommended dose of 300 mg administered on days 1–5 and 8–12 of a 21‐day cycle ...

Adavosertib and beyond: Biomarkers, drug combination and toxicity of WEE1 inhibitors

Critical Reviews in Oncology/Hematology
WEE1 kinase is renowned as an S-G2 checkpoint inhibitor activated by ATR-CHK1 in response to replication stress. WEE1 inhibition enhances replication stress and effectively circumvents checkpoints into mitosis, which triggers significant genetic impairs and culminates in cell death.

Synergistic Effects of Adavosertib, WEE‐1 Inhibitor in Combination with Cytarabine in the Jurkat Leukemia Cell Model

The FASEB Journal, 2022
Current treatment for leukemia focuses on DNA damaging treatments (DNAdt), including chemo (cytarabine, vincristine, doxorubicin, etc.) and/or radiotherapy. However, in both adult and pediatric patients with Acute Lymphoblastic Leukemia (ALL), value of these treatments are limited by development of chemoresistance.

Adavosertib in combination with carboplatin in advanced TP53-mutated platinum-resistant ovarian cancer.

Journal of Clinical Oncology, 2022
5516 Background: Ovarian cancer is globally the second most common cause of death among women with gynecologic malignancies. Despite high initial response rates, the overall prognosis of this patient population remains poor. The majority of advanced ovarian cancers will become platinum resistant defined by recurrence within six months after ...

Single-agent Adavosertib Shows Anticancer Effects Against Colorectal Cancer Cells

Anticancer Research
Colorectal cancer (CRC) is the third most common malignancy and the second most common cause of cancer-related deaths worldwide. Adavosertib (AZD1775), a small molecule inhibitor of WEE1 kinase, abrogates G2/M cell cycle arrest and induces double-stranded DNA breaks.

Abstract B021: PARP16 is a novel target of talazoparib which contributes to synergy with adavosertib in SCLC

Molecular Cancer Therapeutics, 2019
Abstract Background: Small cell lung cancer (SCLC) is a highly aggressive type of neuroendocrine tumor with a low survival rate, signifying an unmet medical need. Although PARP inhibitors (PARPis) display single-agent anticancer activity in SCLC, the underlying biology is poorly understood.

Abstract CT022: A Phase Ib study of Wee1 inhibitor adavosertib in patients with advanced solid tumors

Cancer Research, 2019
Abstract Background: Adavosertib is a highly selective inhibitor of Wee1, a crucial regulator of intra-S and G2/M cell cycle checkpoints. This Phase Ib study (NCT02610075) investigated dosing and schedule for adavosertib monotherapy and determined the maximum tolerated dose (MTD) and recommended Phase II dose (RP2D) in patients (pts ...
