Early vessel destabilization mediated by Angiopoietin-2 and subsequent vessel maturation via Angiopoietin-1 induce functional neovasculature after ischemia. [PDF]
We assessed whether Angiopoietin-2 (Ang2), a Tie2 ligand and partial antagonist of Angiopoietin-1 (Ang1), is required for early vessel destabilization during postischemic angiogenesis, when combined with vascular growth factors.

A Central Somatic Transmission Mediates Proprioceptive Facilitation of Muscle Pain
Zhang et al. uncover a novel central mechanism for persistent muscle pain, in which TRPA1 sensitization in MeV proprioceptive neurons enhances somatic secretion. This, in turn, disinhibits descending pain control from neighboring noradrenergic locus coeruleus neurons via local GABAergic circuits, thereby promoting inflammatory muscle pain.

AAV-based vectors for human diseases modeling in laboratory animals
The development of therapeutic drugs and vaccines requires the availability of appropriate model animals that replicate the pathogenesis of human diseases. Both native and transgenic animals can be utilized as models.

Onset of experimental severe cardiac fibrosis is mediated by overexpression of angiotensin-converting enzyme 2 [PDF]
Angiotensin-converting enzyme (ACE) 2 is a recently identified homologue of ACE. There is great interest in the therapeutic benefit for ACE2 overexpression in the heart.

Hepatocytes are subjected to increased membrane tension along with lipid accumulation. Loss of PIEZO1 exacerbates high‐fat diet‐induced MASLD in mice. Activation of PIEZO1 alleviates high‐fat diet‐induced MASLD in mice. PIEZO1 negatively regulates de novo lipogenesis through activation of CaMKK2‐AMPK pathway. ABSTRACT Liver is a central organ for lipid

Adeno-associated virus for cystic fibrosis gene therapy
Gene therapy is an alternative treatment for genetic lung disease, especially monogenic disorders such as cystic fibrosis. Cystic fibrosis is a severe autosomal recessive disease affecting one in 2500 live births in the white population, caused by ...

Chronic periodontitis elevates circulating CRP, which enters the hippocampus to upregulate BMP4 in oligodendrocyte precursor cells (OPCs), thereby impairing neurogenesis and inducing anxiety/depression‐like behaviors in rats. Counteracting this pathway, CRP deficiency helps confer functional resilience to OPCs.

Mucopolysaccharidosis type IIIB: a current review and exploration of the AAV therapy landscape
Mucopolysaccharidoses type IIIB is a rare genetic disorder caused by mutations in the gene that encodes for N-acetyl-alpha-glucosaminidase. This results in the aggregation of heparan sulfate polysaccharides within cell lysosomes that leads to progressive

This study reveals that abnormal mechanical stress downregulates the expression of HSP70 and impairs proteasome function in osteoarthritic bone cells, leading to misfolded collagen I accumulation and ER stress. When intracellular proteostasis capacity is exceeded, USP19 mediates the secretion of misfolded proteins into the extracellular space ...
