Structure-Activity Studies on Mammalian Adenosine Kinase

Biochemical and Biophysical Research Communications, 2000
The structure-activity relationship for Chinese hamster adenosine kinase (AK) was examined by making systematic deletions from the N- and C-terminal ends. The first 16 a.a. residues from the N-terminal end, which likely form a random coil, can be deleted without any effect on AK activity or stability.

Isolation and Immunological Properties of Adenosine Kinase

Preparative Biochemistry, 1989
Bovine liver adenosine kinase is a 43 kDa protein that catalyzes the transfer of phosphate from GTP or ATP to adenosine. Its immunological properties were compared to other GTP-binding proteins of approximately 40 kDa, in particular those involved in signal transduction, such as Gs and Gi, the stimulatory and inhibitory regulatory proteins of adenylyl ...

Effect of Aging on Myocardial Adenosine Production, Adenosine Uptake and Adenosine Kinase Activity in Rats

Journal of Molecular and Cellular Cardiology, 1999
Adenosine levels present in the interstitial fluid and coronary effluent of the aged heart exceed those of the young adult heart. The present study investigated mechanisms in the Fischer 344 rat heart which may be responsible for the observed differences. (1) Total production of adenosine was determined in isolated perfused hearts by measuring coronary

Modulation of adenosine release from rat spinal cord by adenosine deaminase and adenosine kinase inhibitors

Brain Research, 1995
Adenosine, a modulator of pain processing in the spinal cord, is metabolized by adenosine kinase and adenosine deaminase. In this study we determined which of these mechanisms is more important for the regulation of endogenous adenosine levels in the rat spinal cord.

Adenosine Kinase from Rat Liver: New Biochemical Properties

Nucleosides, Nucleotides and Nucleic Acids, 2006
Adenosine kinase is a well-known enzyme which catalyzes the phosphorylation of adenosine to AMP: Its metabolic and kinetic properties are well studied. Here, we report new properties of rat liver enzyme, demonstrating a new reaction: ADP can be a phosphate donor instead ATP, according to the reaction: adenosine + ADP --> 2AMP) demonstrating the ...

Human Placental Adenosine Kinase: Purification and Characterization

1980
Human placental adenosine kinase has thus been purified 3600-fold and characterized with respect to molecular weight, substrate specificity, divalent cation requirements, pH optimum, isoelectric pH, and kinetic properties. These data contribute to the information currently available about the regulation of adenosine metabolism, information critical for

Adenosine and Deoxyadenosine Kinase from Rat Liver

1980
Recent studies demonstrated that dATP concentration was markedly increased in erythrocytes from patient with adenosine deaminase deficiency1,2 . Deoxyadenosine is toxic to T lymphoblast in the presence of an inhibitor of adenosine deaminase. By the addition of deoxyadenosine, dATP accumulated in the cell and the level of dATP well corelated with the ...
