ADP-ribosylation factor machinery mediates endocytosis in plant cells [PDF]
Endocytosis is crucial for various cellular functions and development of multicellular organisms. In mammals and yeast, ADP-ribosylation factor (ARF) GTPases, key components of vesicle formation, and their regulators ARF-guanine nucleotide exchange factors (GEFs) and ARF-GTPase-activating protein (GAPs) mediate endocytosis.

This study identifies palmitoylation as a novel regulatory modification of SMAD4, mediated by ZDHHC22/APT2. It activates fatty acid synthesis, creating a self‐reinforcing SMAD4–FASN–palmitate feedback loop that drives pancreatic cancer growth and enhances radiotherapy sensitivity.

Control of T lymphocyte morphology by the GTPase Rho [PDF]
Background Rho family GTPase regulation of the actin cytoskeleton governs a variety of cell responses. In this report, we have analyzed the role of the GTPase Rho in maintenance of the T lymphocyte actin cytoskeleton.

Asymmetric nucleosome PARylation at DNA breaks mediates directional nucleosome sliding by ALC1
The chromatin remodeler ALC1 is activated by DNA damage-induced poly(ADP-ribose) deposited by PARP1/PARP2 and their co-factor HPF1. ALC1 has emerged as a cancer drug target, but how it is recruited to ADP-ribosylated nucleosomes to affect their ...

Pancreatic neuroendocrine tumors frequently silence MEN1 through epigenetic mechanisms. Here, SIRT7 recruits DNMT1 to the MEN1 promoter, drives hypermethylation, and enhances DNA repair. Inhibiting SIRT7 restores MEN1, reduces MRN complex abundance, impairs double‐strand break repair, and sensitizes PanNET models to radiation, supporting SIRT7 as a ...

Integrating Spatial Proteogenomics in Cancer Research
Xx xx. ABSTRACT Background: Spatial proteogenomics marks a paradigm shift in oncology by integrating molecular analysis with spatial information from both spatial proteomics and other data modalities (e.g., spatial transcriptomics), thereby unveiling tumor heterogeneity and dynamic changes in the microenvironment.

Are guanine nucleotide binding proteins a distinct class of regulatory proteins? [PDF]
Proteins which bind guanine nucleotides are found in a diverse set of key regulatory positions. They are involved in hormone action, visual transduction, protein synthesis and microtubule assembly. In addition to their ability to bind guanine nucleotides

Opposite Regulatory Effects of Iron Ions on the In Vitro Catalytic Activities of NarE, the Toxin-like ADP-ribosyltransferase from Neisseria meningitidis [PDF]
NarE, the mono ADP-ribosyltransferase identified in Neisseria meningitidis, catalyzes three enzymatic reactions. NarE transfers a single ADP-ribose unit to guanidine compounds, hydrolyses NAD in nicotinamide and free ADP-ribose, and ADP-ribosylates ...
