DNA Methylation Analysis Validates Organoids as a Viable Model for Studying Human Intestinal Aging. [PDF]

open access: yes, 2020
Background & aimsThe epithelia of the intestine and colon turn over rapidly and are maintained by adult stem cells at the base of crypts. Although the small intestine and colon have distinct, well-characterized physiological functions, it remains ...

Loss of stem cell regenerative capacity within aged niches. [PDF]

open access: yes, 2007
This work uncovers novel mechanisms of aging within stem cell niches that are evolutionarily conserved between mice and humans and affect both embryonic and adult stem cells.

The critical role of DNA damage‐inducible transcript 4 (DDIT4) in stemness character of leukemia cells and leukemia initiation

open access: yesMolecular Oncology, EarlyView.
Stemness properties, including quiescence, self‐renewal, and chemoresistance, are closely associated with leukemia relapse. Here, we demonstrate that DNA damage‐inducible transcript 4 (DDIT4) is induced in the hypoxic bone marrow niche and is essential for maintaining the stemness of AML1‐ETO9a leukemia cells.

Potensi Terapi Sel Punca Dalam Dunia Kedokteran Dan Permasalahannya [PDF]

open access: yes, 2008
Stem cell  has become a main focus for not only reseacrhers but also society due to its potency in cell-based therapy. Regardless ethical issues surrounding the human embryonic stem cell, adult human stem cell became the alternative of choice for ...

Evolutionary dynamics of adult stem cells: Comparison of random and immortal strand segregation mechanisms

open access: yes, 2004
This paper develops a point-mutation model describing the evolutionary dynamics of a population of adult stem cells. Such a model may prove useful for quantitative studies of tissue aging and the emergence of cancer.

Nicotinamide N‐methyltransferase promotes drug resistance in lung cancer, as revealed by nascent proteomic profiling

open access: yesMolecular Oncology, EarlyView.
AZD9291 has shown promise in targeted cancer therapy but is limited by resistance. In this study, we employed metabolic labeling and LC–MS/MS to profile time‐resolved nascent protein perturbations, allowing dynamic tracking of drug‐responsive proteins. We demonstrated that increased NNMT expression is associated with drug resistance, highlighting NNMT ...

Therapeutic effects of hypoimmunogenic universal human iPSC-derived endothelial cells in a humanized mouse model of peripheral artery disease

open access: yesStem Cell Research & Therapy
Background Peripheral arterial disease (PAD) is a leading cause of limb disability due to ischemia caused by atherosclerotic plaques. Cell-based therapies using endothelial cells (ECs) have shown promise in promoting angiogenesis for PAD, but challenges ...
